摘要
利用自主开发研制的作物脂肪氧化酶同功酶快速检测技术 ,筛选出Lox -1、Lox -2缺失材料皖鉴 2 0 90、皖鉴 10 2 4、皖鉴3 0 3 3等。为了进一步研究鉴定脂质氧化酶缺失对稻谷陈化、仓储害虫的影响 ,将 3个Lox缺失材料 (DawDam、冲腿、皖鉴 2 0 90 )和其他 3个Lox不缺失品种 (早籼 14、中籼 898、晚粳M3 12 2 ) ,在一定的储藏条件、温度、时间下 ,调查了仓储害虫危害情况和种子生活率。结果表明 :①在储藏过程中 ,品种间仓储害虫危害差异较大。在 5个籼稻品种中 ,Lox -3缺失种质DawDam和冲腿的虫蛀率约 5 % ,为最低 ;中籼 898、皖鉴 2 0 90、早籼 14的虫蛀率在 13 .8%~ 19.5 % ;粳稻品种晚粳M3 12 2的虫蛀率最高 ,达 3 3 .5 %。因此 ,Lox -3缺失种质的虫蛀率的发生较其他品种低 2 .5~ 6.5倍。②随着储藏期的增加 ,种子发芽率呈下降趋势。但是Lox -1、Lox -2缺失的皖鉴 2 0 90 ,至调查时储存 42个月 ,发芽率几乎没有变化 ,仍保持在 98.5 % ,成苗率高达 85 %。由此表明 ,Lox -1、Lox -2的缺失对保持种子生活力和延缓稻谷陈化变质具有重要的作用 。
Rice materials wanjian?2090,wanjian?1024,wanjian?3033 with the loss of Lox-1 and 2 were screened with the patent technique of Lox isoenzymes detective.In order to study the effects of Loxs loss on rice deterioration and storage insects, three rice materials with the loss of Loxs: Daw Dam,Chongtui,Wanjian 2090 and other three normal rice varieties:Earlier indica rice?14,Media indica rice?898,Later japonica rice M3122 without the loss of Loxs were respectively packed and storied in same storage conditions. The degree of rice grain damaged by insects and seed viability was respectively investigated during storage period. The results were as follows:①There was a significant difference in the degree of rice grain damaged by insects. In ratio of insect injury: Both Daw Dam and Chongtui with the loss of Lox-3 were about 5%,which was the lowest, Middle indica rice?898,Wanjian?2090,Earlier indica rice?14 were among 13.8%~19.5%,Later japonica rice M3122 were highest at 33.5%. So, the ratio of insect injury of materials with the loss of Lox-3 was 2.5 to 6.5 times lower than that of materials without the loss of Lox-3.②With the increase of storage time, the seed germination of most of materials were decreased. But the germination of Wanjian?2090 with the loss of Lox-1 and 2 was still as high as 98.5% which had little change after 42 months' storage.The rice material with the loss of Lox-1 and 2 was very important in its seed viability, and Lox-1,Lox-2 could be the key genes that affect the seed long span.
