以成龄巴西橡胶树为材料,用免疫印迹技术研究了胶乳中的肌动蛋白,并用异硫氰酸四甲基罗丹明标记的鬼笔环肽荧光探针检查了乳管伤口的肌动蛋白细胞骨架。割胶后收集的胶乳,经超速离心分离的各个组分中,C乳清(胞质溶液)组分含有肌动蛋白。排胶初始期流出的胶乳中肌动蛋白含量多,排胶后期流出的胶乳中肌动蛋白的含量减少。比较了不同割胶强度下收集的胶乳中肌动蛋白的含量,结果表明在强度割胶条件下,胶乳中的肌动蛋白含量较少。用显示肌动蛋白微丝的鬼笔环肽荧光探针检测到,停止排胶时的乳管伤口末端有一团呈现橙红色的荧光物质,表明了肌动蛋白微丝在伤口末端聚积。进行了外施肌动蛋白微丝的解聚剂大环内脂和碘化钾刺激采胶试验,结果表明:施用质量分数1%的碘化钾和饱和浓度的大环内酯水溶液能使胶乳产量增加。这些事实表明,肌动蛋白细胞骨架可能在橡胶树乳管排胶和乳管堵塞中起重要作用。
Latex from adult trees of Hevea brasiliensis Mull. Arg. was tested for actin by using immunobloting with actin antiserum, and the actin cytoskeleton in laticifer wound was investigated by using TRITC Phalloidin fluorescent optical microscopy. Actin was detected in c serum (cytosol) of the latex fractions resulted from ultra-centrifugation. More actin was found in the latex collected immediately after tapping than in the latex collected before latex flow stopped. A comparison between trees with varied tapping intensities showed that less actin appeared in the latex from the trees with more intensive latex exploitation. A mass of TRITC-fluorescent substance was detected in laticifer wounds when latex flow stopped, indicating accumulations of actin cytoskeleton in the wounds. Stimulated exploitation of latex was made with macrolides and KI,which are depolymerizing compounds of actin cytoskeleton. The results showed that application of saturated macrolides solution in water or 10 g/kg KI led to increase in latex yield. The results of the present stndy indicate that actin cytokeleton might play an important role in regulation of latex flow and wound plugging of the laticifers in Hevea brasiliensis.
