半枝莲多糖的分离、纯化及其理化性质 被引量:31

ISOLATION AND PHYSICO-CHEMICAL PROPERTIES OF POLYSACCHARIDE FROM THE HERB OF SCUTELLARIA BARBATA D.DON

摘要 唇形科植物半枝莲全草的热水提取液经乙醇沉淀、去蛋白、逆向流水透析、DEAE-纤维素柱层析和Sephadex G-100柱层析得纯品半枝莲多糖(S-BP)。凝胶过滤及聚丙烯酰胺凝胶电泳证明为单一均匀成分。SBP的比旋度为[α]_D^(20)=+26.2°,不含氮,分子量13000,具有890cm^(-1)红外吸收峰。经纸层析和气相色谱分析,SBP含鼠李糖、阿拉伯糖、木糖、甘露糖、半乳糖和葡萄糖,其克分子比为1.14:1.50:0.20:0.75:1.0:2.18。 SBP, a polysaccharide component of scutellaria barbata D. Don was prepared by ethanol precipitation of water extract of the herb, that pre- liminarily extracted by ethanol, followed by removal of proteins, dialysis against water, precipitaion with alcohol and further purification by ch- romatography on DEAE-cellulose column and sephadex G-100 column. SBP is shown to be homogencous on gel permeation and polycrylamidegel electrochromatophoresis. SBP showed positve optical rotation: [α]_D^()20=+ 26.2 (C=1.2, H_2O). The IR spectrum of SBP had an absorption band at 890cm^(-1), due to a β-glycoside linkage. Its number-average molecular weight was estimated to be 13000. By PC and GC, SBP was showed to be composed of Rham, Ara, Xy1, Man, Ga1, G1u, and its molar ratio is 1.14:1.5:0.2:0.75:1.0:2.18.
