摘要
目的 探讨登革病毒 2型 (DEN2 )临床分离株感染BALB c小鼠后其血浆中IL 4、IFN γ产生动态及相关关系。方法 用不同剂量DEN2临床分离株经皮下多点注射 ,建立BALB c小鼠感染模型 ,并用双抗体夹心ELISA法检测感染后不同时间各组动物血浆中IL 4、IFN γ浓度。结果 DEN2感染可使BALB c小鼠产生IL 4、IFN γ增加 (P <0 .0 5 ) ,且细胞因子浓度与病毒感染剂量有关。初次感染阶段两者水平均有升高 ,而再次感染阶段以IL 4升高为主。结论 在初次感染阶段TH1 TH2应答均发挥重要作用 ;而再次感染阶段以TH2应答为主。
Objective To observe the dynamic levels of IL-4 and IFN-γ produced by BALB/c mice infected with Dengue virus type Ⅱ (DEN2) clinic strain and to study their relationship. Methods The BALB/c murine infection model was established by multiple-site subcutaneous injection with various doses of DEN2 clinic strain. Murine plasma samples collected from different experiment groups at various time after infection were tested for IL-4, IFN-γ levels with sandwich ELISA. Results The BALB/c mice increasingly produced IL-4 and IFN-γ after DEN2 infection. The concentrations of the cytokines produced by infected animals were closely linked to the dosage of DEN2. Both levels of IL-4 and IFN-γ increased after the initial infection, but IL-4 increased more predominantly than IFN-γ in the reinfection phase. Conclusion Both T_H1 and T_H2 responses were induced by DEN infection in mice in the initial infection phase; while T_H2 response was more predominant than T_H1 response in the second infection phase.
