摘要
目的 :通过干扰体外培养的成骨样细胞 ,探索糖皮质激素所致成骨样细胞凋亡的分子细胞机制。 方法 :地塞米松诱导原代培养并传代的小鼠成骨样细胞 ,应用噻唑蓝 (MTT)法检测细胞生存能力 ,流式细胞仪检测细胞凋亡 ,比色法检测半胱天冬酶Caspase 3的活性以及EMSA检测核因子KappaB(NF κB)活性。 结果 :地塞米松使成骨样细胞存活数量下降 ,Caspase 3活性升高 ,NF κB活性下降。 结论 :地塞米松对原代未转化的成骨细胞有诱导凋亡的作用 ,并且是Caspase 3依赖性的 ,但NF κB似乎不参与该过程。
Objective:To explore cytological and molecular mechanism of osteoblast apoptosis induced by glucocorticoids using osteoblast-like cells cultured in vitro. Methods:Dexamethasone was used to induce primary cultured and subcultured osteoblast-like cells from mouse. MTT, FCM, colorimetry and EMSA were used to define survival of the cells, apoptosis, activity of Caspase-3 and NF-κB respectively.Results:Dexmethasone increased the apoptosis of osteoblast-like cells, induced the activity of Caspase-3 while inhibited the activity of NF-κB. Conclusion:Dexamethasone is capable of inducing the apoptosis of primary cultured and non-transformed osteoblasts, which is Caspase-3 dependent, while NF-κB doesn’t seem to participate in the procedure.
