摘要 目的 :探讨诱导型一氧化氮合酶 (iNOS)在大鼠视网膜缺血再灌注损伤中的表达及意义。方法 :建立结扎颈总动脉的大鼠视网膜缺血再灌注损伤模型 ,6 0只SD大鼠被随机分为缺血再灌注组和对照组。每组按再灌注后不同时间段相应分为 1h、6h、12h、2 4h、4 8h、72h小组 ,每小组 5只大鼠 ,以免疫组织化学法 (SABC法 )检测视网膜组织中iNOS的表达情况。结果 :在缺血再灌注损伤 1h时 ,未见到iNOS的表达 ,在缺血再灌注 6h时 ,iNOS开始表达 ,至第 2 4小时表达最强 ,第 4 8小时开始减弱 ,但第 72小时仍有表达。而对照组视网膜中未能检测到iNOS的表达。结论 :iNOS可能在视网膜缺血再灌注损伤中发挥重要作用。 Objective:To study iNOS expression in the rat retina that has been injured by ischemia-reperfusion.Methods:A retina model for ischemia-reperfusion was established by ligation of the common carotid artery in the SD rat. 60 SD rats were divided into two groups. Each group was then divided into six sub-groups according to differences in ischemia-reperfusion time. The SABC method was used to test iNOS expression in the rat retina.Results:At one hour after ischemia-reperfusion,no iNOS was expressed in the retina. After six hours of ischemia-reperfusion,there was some degree of iNOS expression in the retina. The highest level of expression was reached at twenty four hours after ischemia-reperfusion. The level then began to decrease,but there was evidence of iNOS expression in the retina seventy-two hours after ischemia-reperfusion injury.Conclusion:iNOS may play a very important role in ischemia-reperfusion injury in the rat retina.
