摘要
目的:观察三七总甙对体外培养的人瘢痕疙瘩成纤维细胞增殖及胶原合成的作用,以寻找治疗人瘢痕疙瘩的有效药物。方法:体外培养人瘢痕疙瘩成纤维细胞,分别应用噻唑蓝(MTT)比色法和H3-脯氨酸掺入法检测三七总甙作用后细胞增殖及胶原合成的变化。结果:与空白对照组相比,三七总甙含量在0.5,1.0和1.5g/L时均能够显著抑制成纤维细胞增殖及胶原合成(P<0.01),但在1.5g/L时对细胞具有毒性作用。结论:三七总甙具有体外抗纤维化作用,可能成为治疗瘢痕疙瘩的有效药物。
AIM:To observe the effect of panax notogin seng on the proliferation and collagen synthesis of the cultured human keloid fibroblasts in vitro and seek the effective drug for human keloid. METHODS:The human keloid fibroblasts were cultured in vitro.The thiazolyl blue MTT chromatometry test and H3 proline incorporation method were used to detect the effect of panax notogin seng on the proliferation and collagen synthesis of the cultured human keloid fibroblasts. RESULTS:When compared with the control groups,panax notogin seng could markedly inhibit the proliferation and collagen synthesis of the cultured human keloid fibroblasts at the concentration of 0.5,1.0 and 1.5 g/L (P< 0.01),but has toxic effect on them at the concentration of 1.5 g/L. CONCLUSION:Panax notogin seng has anti fibrotic effect in vitro and may be an effective drug for human keloid.
