摘要
目的:优选桔梗茎叶总皂苷的最佳提取和纯化工艺,为桔梗非药用部位的综合开发利用提供参考。方法:以桔梗茎叶总皂苷收率、桔梗皂苷D的含量与转移率为指标,采用正交试验考察溶剂用量、提取温度、提取时间与提取次数对桔梗茎叶皂苷提取工艺的影响,再进一步通过大孔吸附树脂富集纯化总皂苷。结果:最佳提取工艺参数为在100℃条件下,加12倍量的水,提取2次,每次1.5 h。优化纯化工艺参数为最佳工艺提取药液离心后浓缩至1.0 g生药/mL,通过AB-8型大孔吸附树脂,上样量为桔梗茎叶提取物量∶树脂量=0.8∶1,上样流速为1.0 mL·min^(-1),用5 BV的75%乙醇洗脱。结论:优选的提取和纯化工艺稳定可行,可用于桔梗茎叶皂苷的提取。
Objective:To establish a suitable extraction and purification process of the total saponins from stem and leaf of Platycodon grandiflorum,in order to lay a foundation for the comprehensive utilization of the non-medicinal parts of P.grandiflorum.Methods:With the yield of total saponins,the content and transfer rate of saponin D as index,the effect of extraction was investigated.The extraction conditions included solvent amount,extraction temperature,extraction time and extraction times.The total saponins were further enriched and purified by macroporous adsorption resin.Results:The optimized extraction conditions were extraction temperature of 100℃,extraction 2 times,each time 1.5 hour and solid-to-liquid ratio of 1∶12(g·mL-1).The parameters of the optimized purification process:centrifuged and concentrated the soup extracted through the optimal process to 1.0 g·mL-1,the extracts were adsorbed with AB-8 macroreticular resin and then eluted by 75%alcohol.Extraction amount∶resin amount=0.8∶1,the flow rate was 1.0 mL·min-1.Conclusion:The optimal extraction and purification process is stable and easy,and can be used in the extraction of total saponins from stem and leaf P.grandiflorum.
