摘要
用几种蛋白质侧链修饰试剂对β-N-乙酰氨基己糖苷酶进行化学修饰,在一定条件下,当巯基、羟基、酪氨酸残基分别被 IAA 及 NEM、PMSF、NAI 修饰后,酶活力不受影响,说明这些基团与活力无关。当羧基、组氨酸及色氨酸残基分别被 EDC、DEP、NBS 修饰后,酶活力大幅度下降,说明这些基团或者参与了酶催化作用,或者位于酶活性位区附近。
The effects of some protein modifica-tion reagents on the β-N-Acetylhexosamini-dase activity have been studied.The enzymewas not affected by IAA and NEM,PM-SF,NAI modification thereof,suggestingthat thiol group,hydroxyl group and tyro-sine residues were non-essential to enzymeactivity.But the enzyme activity was remar-kably diminished after EDC,DEP,NBS mo-dification.These results indicated that thecarboxyl groups,histidine and tryptophanresidues seemed to be essential to catalyticactivity or located at the active site of theβ-N-Acetylhexosaminidase.
