摘要 目的优选超临界CO2流体萃取虎杖中有效成分白藜芦醇的最佳工艺条件。方法采用均匀设计法安排试验,探索压力、温度、夹带剂等工艺参数对有效成分得率的影响,采用高效液相色谱(HPLC)法测定虎杖萃取产物中白藜芦醇的含量,计算收率,数据处理采用SAS 8.0统计软件处理。结果最佳萃取条件为萃取压力30 MPa,萃取温度60℃,夹带剂用量为100%,萃取时间120 min。结论超临界CO2流体萃取较常规的有机溶剂提取方法可获得更高的收率,为改进虎杖中白藜芦醇的提取方法提供了科学依据。 Objective To optimize the optimal extraction conditions of the effective constituent resveratrol in polygonum cuspidatum by CO2 supercritical fluid extraction( SFE). Methods The uniform design method was used to arrange the experiment for investigating the influence of the technological parameters of the pressure,temperature and entrainer on the yield rate of the effective constituent. The HPLC method was adopted to determine the resveratrol content in the polygonum cuspidatum extraction product. The yield rate was calculated and the data were processed by SAS 8. 0 statistical software. Results The best extraction conditions by SFE were as follows:the extraction pressure 30 MPa,extraction temperature 60 ℃,the entrainer amount 100% and the extraction time 120 min. Conclusion The CO2 SFE method acquires more yield than the conventional organic solvent extraction method and provides the scientific basis for improving the extraction method of resveratrol.
