摘要
目的研究决明子的高效液相色谱(HPLC)特征图谱,为科学评价省评价性抽验品种决明子的质量现状提供依据。方法特征图谱采用Waters Sun Fire TM C18(250 mm×4.6 mm,5μm)色谱柱,以乙腈-磷酸溶液为流动相梯度洗脱。色谱数据采用相似度评价软件分析并对结果进行聚类分析,探讨特征图谱与质量的相关性。结果分别建立了决明及炒决明的HPLC特征图谱;聚类分析结果表明,市场上的决明子质量参差不齐,相似度与现行药典标准含量测定合格率有很大相关性。结论决明子总体质量较差,质量标准有待商榷。建立的特征图谱能清晰反映决明子生品和炮制品的真实质量,可用于该药的质量控制。
Objective To study the HPLC fingerprint of Semen Cassiae for its scientific quality evaluation. Methods Waters Sun Fire TM C18 column( 250 mm × 4. 6 mm,5 μm) was adopted with acetonitrile- phosphoric acid water was treated as mobile phase in gradient elution; the chromatogram was analyzed by the traditional Chinese medicine( TCM) fingerprint similarity evaluation software and the similarity result was analyzed by the similarity clustering system. The correlation between fingerprint and quality was explored. Results The fingerprints of Semen Cassiae and its processed products were set up respectively. Cluster analysis results showed the quality in the market had significant difference,the similarity had great relevance with the percent of pass detected according to the current pharmacopoeia standard. Conclusion The overall quality of Semen Cassiae is poor,the quality standards remains open to question. The newly founded fingerprint method can clearly reflect the actual quality of Semen Cassiae and its processed product,and can be used for the quality of Semen Cassiae.
