摘要
目的 :研究HBx蛋白对细胞凋亡的调节及其在肝细胞癌发生中的作用。方法 :采用SP免疫组化方法和原位脱氧核糖核酸末端转移酶标记技术分别检测HCC组织中HBx和p5 3蛋白表达及细胞凋亡。结果 :HCC和癌旁组织中的HBx蛋白的阳性率分别为 5 2 5 % (2 1 4 0 )和 6 7 5 % (2 7 4 0 ) ,在HCC中阳性信号多为呈核型 ,癌旁组织中以核浆型为主 ,阳性细胞多呈区域性或簇状分布 ;HCC中p5 3和HBx蛋白的表达具有较高的一致性 (P <0 0 1) ;p5 3和HBx表达强度均与凋亡细胞相关 ,其表达增强则凋亡细胞密度逐渐降低 (P <0 0 1)。结论 :提示HBx蛋白可能通过抑制p5 3的功能阻止细胞凋亡和促进细胞增殖 。
Objective To study the regulation of HBx protein to apoptosis and their effect in hepatocelular carcinogenesis.Methods The expressions of HBx and p53 proteins were detected by SP immunohistochemistry and the apoptosis by in situ terminal deoxyribonucleotide transferase labeling.Results The positive rates of HBx protein were 52.5%(21/40) and 67.5% (27/40) in HCCs and their surrounding liver tissues respectively.The positive signal of HBx protein in HCC was mainly in nuclei,and that in pericarcinomatous liver tissues was nuclear-cytoplasm type.The distribution of the positive cells was regional or clustered.There was better concordance between expressions of HBx and p53 protein in HCCs.The expressions of HBx and p53 protein were related to apoptosis,the stronger expression of them,the lower density of apoptotic cells in HCCs.Conclusions The data suggest that HBx protein may prevent apoptosis and accelerate cell proliferation,and result in uncontrolled cell proliferation and more malignant phenotype by suppressing p53 protein function in hepatocellular carcinogenesis.
