摘要
目的:探讨大黄素抑制同种异体小鼠皮肤移植排斥反应的作用。方法:采用BALB/c供体鼠→C57BL/6受体鼠背-背皮肤移植法建立模型,分为假手术组、模型组、大黄素组(50 mg/kg)、环孢素A组(10 mg/kg)、大黄素+亚剂量环孢素A组(大黄素50 mg/kg+环孢素A 5 mg/kg),腹腔注射给药10 d.记录受体鼠移植皮片的存活时间;取血浆,酶联免疫吸附法检测Th1类细胞因子(IL-2,IFN-γ)和Th2类细胞因子(IL-4,IL-10)水平;取脾脏,流式细胞仪分析T淋巴细胞亚群CD4^+及CD25^+的比例。结果:与模型组比较,各给药组受体鼠移植皮片的存活时间均有明显延长,组间差异均有统计学意义(P<0.01);大黄素联合环孢素A组的移植皮片存活时间比大黄素组或环孢素A组均有明显延长,差异有统计学意义(P<0.05)。与模型组比较,大黄素联合环孢素A组及大黄素组均可显著降低血浆IL-2和IFN-γ的水平,差异有统计学意义(P<0.01),显著升高IL-4和IL-10的水平,差异有统计学意义(P<0.01)。与模型组比较,大黄素联合环孢素A组及大黄素组均可显著升高脾脏CD4^+及CD25^+调节性T细胞的比例,差异有统计学意义(P<0.01)。结论:大黄素可抑制同种异体小鼠的皮肤移植排斥反应,其机制或与促使Thl向Th2细胞的偏移,刺激CD4^+及CD25^+调节性T细胞的表达,诱导免疫耐受等相关。
Objective:To investigate the effects of emodin against mice with xenogenic skin transplantation.Methods:The tergal skin transplantation model was achieved by an operation(BALB/c mice as donors and C57 BL/6 mice as acceptors,respectively).Then acceptors were divided randomly into 5 groups,namely sham,model,emodin(50 mg/kg),ciclosporin A(10 mg/kg),combination of emodin and ciclosporin A(emodin 50 mg/kg and ciclosporin A 5 mg/kg).All acceptor mice were intraperitoneally administered 10 d.The skin graft survival time of all mice were documented every day.At the end of the experiment,the levels of Th1 cytokines(IL-2 and IFN-γ)and Th2 cytokines(IL-4 and IL-10)in plasma were detected by ELISA and the spleen in all mice were removed and quantity of CD4^+,CD25^+T cells were analysed by flow cytometry.Results:Compared with the survival days of skin graft of model group,those in groups treated with drugs were all significantly prolonged(P<0.01).Combination of emodin and ciclosporin A group showed a longer survival days than emodin or ciclosporin A group(P<0.05).IL-2 and IFN-γlevels in plasma in groups treated with emodin or combination of emodin and ciclosporin A were evidently decreased(P<0.01)while IL-4 and IL-10 levels were evidently increased(P<0.01)comparing with those of model group.Compared with the amount of CD4^+,CD25^+T cells in spleen of model group,those in groups treated with emodin or combination of emodin and ciclosporin A were all significantly increased(P<0.01).Conclusion:Emodin has an immunosuppressive effect against mice with xenogenic skin transplantation.The effectiveness of emodin appears to be associated with balance of Th1/Th2 cytokines and increasement of expression of CD4^+,CD25^+T cells,etc.
