摘要
综合运用硅胶柱、MCI以及制备HPLC等多种色谱技术对青龙衣95%甲醇提取物进行分离纯化,利用NMR,MS等波谱技术结合理化性质进行结构鉴定,从青龙衣95%甲醇提取物的二氯甲烷萃取部位中分离鉴定出了14个化合物,分别为5-hydroxy-4-methoxy-1-tetralone(1)、4,8-dihydroxy-1-tetralone(2)、4,5-二羟基-α-四氢萘醌(3)、blumenol B(4)、dehydrovomifoliol(5)、megastigm-5-ene-3,9-diol(6)、胡桃宁B(7)、blumenol C(8)、黑麦草内酯(9)、oleracone B(10)、丁香脂素(11)、松脂醇(12)、香草酸甲酯(13)和异香草酸(14)。其中化合物1,4~6以及8~13为首次从青龙衣中分离得到。化合物4~6,8,10,12和13为首次从胡桃属中分离得到。采用MTT法对14个化合物针对子宫颈癌细胞(HeLa)、人胃癌细胞(HGC-27)、结肠癌细胞(Ht-29)3种肿瘤细胞株的体外肿瘤细胞抑制活性进行评价。结果表明化合物2,3,7,9,11对3种肿瘤细胞株具有不同的抑制活性,其中化合物7活性较强,其IC50分别为26.5,9.0,25.4μmol·L-1。
Fourteen chemical constituents,including 5-hydroxy-4-methoxy-1-tetralone(1),4,8-dihydroxy-1-tetralone(2),4,5-dihydroxy-α-tetralone(3),blumenol B(4),dehydrovomifoliol(5),megastigm-5-ene-3,9-diol(6),juglanin B(7),blumenol C(8),loliolide(9),oleracone B(10),syringarsinol(11),pinoresinol(12),methyl 4-hydroxy-3-methoxybenzoate(13),and isovanillic acid(14),were isolated from the dichloromethane fraction of 95%methanol extract of green walnut husks by silica gel and MCI column chromatography,and Pre-HPLC.Their structures were determined by spectroscopic methods,such as NMR,MS and so on.Among them,compounds 1,4-6,8-13 were isolated from the green walnut husks for the first time,and compounds 4-6,8,10,12,13 were isolated from the Juglans genus for the first time.All of isolates were detected their inhibitory activities against HeLa,HGC-27 and Ht-29 cell lines by the MTT assay.The result showed that compounds 2,3,7,9 and 11 exhibited inhibitory activity against the tested cell line.The IC50 of 7 were 26.5,9.0,25.4μmol·L-1,respectively.
