摘要
目的:建立人胚胎嗅神经鞘细胞(OECs)的体外培养体系.方法:选取3~6个月的流产儿,采用原代培养的方法,在原代培养后的不同时间,用组织免疫化学染色的方法鉴定细胞并计算阳性率.找到细胞数量和阳性率俱佳的时间段,为以后的移植工作打下基础.结果:成功培养出人胚胎OECs,在原代培养后的2~5d主要为巨噬细胞状、多极状,与成纤维细胞很难分别,7~15d主要为扁平的双极、三极形态,突起细长,细胞阳性率可达90%,21d以后为双极、三极形态.结论:自胚胎培养OECs的方法实用可行,方法简单,为利用OECs修复脊髓损伤进入临床提供了条件.
Obje ctive:To set up the culture system of olfactory ensheathing cells in vitro from human em-bryo.Method:Choosing three to six months old abortus for primary cultu re,and during the specific culture peri-ods the cells were evaluated by means o f immunochemistry staining and the positive rate was calculated as well.The suit able period of time when the cells with the most quantity and the best proportio n of positive was found for the following employment.Result:The OECs were cultur ed successfully with good feasibility and re-peatability.In2to5days cells exhib ited macrophage,multipolar which were difficult to differ from fibroblast.In7to1 5days the majority of cells became flat bipolar,tripolar with90percent of positi ve.After21days they appeared as bipolar and tripolar with poor proliferation and vitality.Conclusion:Culturing OECs from embryo is feasible and it can provide a condition for restoring spinal cord injury by using OECs.
