摘要
碳化二亚胺法将伏马菌素B1(fumonisinB1,FB1)与载体蛋白进行偶联制备免疫抗原FB1-BSA及包被抗原FB1-OVA。紫外扫描初步显示载体蛋白与伏马菌素偶联成功。皮下注射免疫BALB/c小鼠,获抗FB1多克隆抗体,间接非竞争ELISA分析所得抗血清效价,最高为1∶10000以上,间接竞争ELISA显示所制备的抗血清能有效识别伏马菌素B1,与脱氧雪腐镰刀菌烯醇(DON)、玉米赤霉烯酮(ZEN)、T-2毒素无交叉反应。利用自制抗体建立啤酒中FB1间接竞争ELISA检测方法;对啤酒5个加标浓度的回收率在71.88%～106.93%。
EDC method was used to conjugate fumonisin B1 to carrier protein,then artificial antigen and carrier protein were respectively taken on UV spectrum scan.The result showed that artificial antigen was synthesized successfully primarily.BALB/c small mice were immunized with immunogen(FB1-BSA)through subcutaneous,the titer of the anti-fumonisin B1 antibodies were detected by indirect ELISA,the highest titers of the antisurem was higher than 1:10000,indirect competive ELISA showed that the polycloal anti-fumonisin B1 andibodies could detect fumonisin B1 specificilly,there was no cross reaction with DON,ZEN and T-2 toxin.Enzyme-linked immunosorbant assay for detection of FB1 in bear was developed using the polycolnal antibodies.The recovery rate of beer on the level of 5ng/ml,50 ng/ml,100 ng/ml,200 ng/mland 500 ng/ml was among 71.88%～106.93%.
