摘要
重组痘苗病毒候选疫苗rVTT-JEVE投入使用之前需要进行本体动物免疫和攻毒保护试验,本研究将重组痘苗病毒rVTT-JEVE进行仔猪免疫接种,分别在体液免疫及细胞免疫水平上评价其免疫效果,并检测攻毒后仔猪体内乙型脑炎病毒载量,分析讨论乙型脑炎重组痘苗病毒候选疫苗rVTT-JEVE对猪体的免疫效果。经2次仔猪免疫接种后,重组痘苗病毒rVTT-JEVE组的IgG抗体水平是商品疫苗SA-14-14-2组的1.5倍,显著高于SA-14-14-2组(P<0.05),而且rVTT-JEVE组的中和抗体效价达到SA-14-14-2组的1.7倍,显著高于SA-14-14-2组(P<0.05)。rVTT-JEVE组的T淋巴细胞刺激指数为SA-14-14-2组的1.6倍,显著高于SA-14-14-2组(P<0.05)。本研究结果显示,rVTT-JEVE候选疫苗组和SA-14-14-2商品疫苗组JEV病毒载量均显著低于PBS对照组(P<0.05),均可有效保护仔猪感染JEV。
Recombinant vaccinia virus candidate vaccine rVTT-JEVE needs to carry out ontology animal immunology and protection research challenged with JEV before used.In this study,the recombinant vaccinia virus rVTT-JEVE was used for immunization in piglets.The immune effects of rVTT-JEVE were evaluated at humoral and cellular levels,and viral loads in piglets after challenged with JEV.After immunized twice in piglets,the IgG antibody level of the recombinant vaccinia virus rVTT-JEVE group was 1.5times higher than that the licensed vaccine SA-14-14-2 group(P<0.05).The neutralizing antibody titer of the rVTT-JEVE group was 1.7-fold higher than that of the SA-14-14-2group(P<0.05).T-lymphocyte stimulation index of rVTT-JEVE group was 1.6times higher than that of SA-14-14-2group(P<0.05).The results showed that the viral load of JEV in the rVTT-JEVE candidate vaccine group and the SA-14-14-2licensed vaccine group were significantly lower than that in the PBS control group(P<0.05),and both of them could effectively protect the piglet from infecting JEV.
