饲粮添加陈化玉米和复合抗氧化剂对仔猪空肠屏障及后肠微生物和代谢产物的影响 被引量:3

Effects of aging corn and complex antioxidant supplementation on the jejunal barrier and hindgut microbes and metabolites of piglets

摘要 选取体质量为(7.24±0.02)kg的健康'杜洛克×长白×约克夏'(DLY)断奶仔猪24头,按体质量相近和公母各半的原则随机分为4个处理,每个处理6个重复,每个重复1头猪,分别饲喂基础饲粮、陈化玉米50饲粮(陈化玉米替代基础饲粮玉米的50%)、陈化玉米100饲粮(陈化玉米100%替代基础饲粮玉米)、复合抗氧剂饲粮(陈化玉米100饲粮+0.2%复合添加剂)。试验期28 d。分别考察仔猪血清免疫指标、空肠屏障基因及后肠微生物和代谢产物等相关指标。结果显示:(1)添加陈化玉米及复合抗氧化剂对仔猪血清先天性免疫指标无显著影响(P>0.05)。(2)与基础饲粮组相比,添加陈化玉米显著升高血清中TNF-α的水平(P<0.05),显著降低抗炎因子IL-10 mRNA表达丰度(P<0.05),对其他相关炎性和凋亡相关指标无显著影响(P>0.05)。(3)与基础饲粮相比,添加陈化玉米显著降低空肠GSH-Px的活力(P<0.05),且陈化玉米100组显著提高空肠PCO水平(P<0.05);同时,陈化玉米50组和陈化玉米100组仔猪血清CAT酶活分别降低26%和35%,且陈化玉米100组T-SOD活性降低10.6%,GSH含量下降21.99%,但差异均不显著(P>0.05);与陈化玉米100组相比,添加复合抗氧化剂显著降低脂质过氧化产物MDA水平(P<0.05)。(4)添加陈化玉米和复合抗氧化剂对空肠黏膜紧密连接蛋白基因表达水平均无显著影响(P>0.05)。(5)与基础饲粮相比,陈化玉米100组显著提高空肠黏膜SGLT1的mRNA表达丰度(P<0.05);与陈化玉米100组相比,添加复合抗氧化剂对空肠养分转运载体的表达水平无显著影响(P>0.05)。(6)添加陈化玉米和复合抗氧化剂对空肠、盲肠、结肠食糜pH值,以及盲肠、结肠微生物数量及短链脂肪酸含量均无不显著影响(P>0.05)。结果表明,陈化玉米不同比例替代普通玉米一定程度诱导机体炎症反应、降低空肠抗氧化能力,但不损伤肠道屏障功能,且可能引发部分肠道黏膜养分转运载体基因表达上调。 A total of 24 healthy weaned Duroc×Landrace×Yorkshire(DLY)piglets,average(7.24±0.02)kg,were randomly divided into 4 treatments with 6 replicates per treatment and 1 piglet per replicate,which were fed basal diet,aging corn 50 diet(50%normal corn in basal diet was substituted by the same amount of aging corn),aging corn 100 diet(100%normal corn in basal diet was substituted by the same amount of aging corn),complex antioxidants diet(aging corn 100 diet+0.2%complex antioxidants).The trial lasted 28 d.The immune indexes,intestinal mechanical barriers,microbial barriers,and metabolites of the hindgut in piglets were measured separately.The results showed as follows:(1)There was no significant effect on serum innate immune index of piglets by supplementation of aging corn and complex antioxidants(P>0.05).(2)Compared with the basal diet group,the aging corn group significantly increased serum TNF-αlevel(P<0.01),and significantly reduced the jejunal IL-10 mRNA expression abundance(P<0.05).It didnt display significant effect on other related inflammatory and apoptotic indexes(P>0.05).(3)Compared with the basal diet group,the aging corn group significantly reduced the activity of GSH-Px in jejunum(P<0.05),even more the aging corn 100 group significantly increased the level of PCO(P<0.05).Meanwhile,the CAT activities of aging corn 50 group and aging corn 100 group respectively decreased by 26%and 35%,also the aging corn 100 group decreased the activity of T-SOD in 10.6%,and the content of GSH in 21.99%,but no significant effect(P>0.05).Compared with the aging corn 100 group,complex antioxidants group significantly reduced the level of MDA(P<0.05).(4)Addition of aging corn and complex antioxidants did not significantly affect the mRNA expression level of the tight junction protein in jejunal mucosa(P>0.05).(5)Compared with the basal diet,aging corn 100 group significantly increased the SGLT1 mRNA expression in jejunal mucosa(P<0.05).Compared with aging corn 100 group,Supplementation with complex antioxidants did not significantly affect the expression of jejunal nutrient transporter(P>0.05).(6)Addition of aging corn and complex antioxidants did not significantly affect the pH value change of jejunum,cecum,and colon chyme,and the number of microorganisms and short-chain fatty acids in the cecum and colon(P>0.05).It can be seen that different proportions of aging corn replaced common corn could induce the body’s inflammatory reaction and reduce the oxidative capacity of jejunum.However,it does not damage the intestinal barrier function and may leads to the up-regulation of part of intestinal mucosal nutrient transporter gene expression.
