摘要
目的:旨在测定不同烘制温度下党参炮制品中党参炔苷的含量,为科学评价及有效控制其质量提供可靠方法。方法:采用HPLC法、梯度洗脱及超声提取法。色谱条件为:色谱柱:Kromasil C18色谱柱(4.6 mm×250 mm,5μm);流速:1.0 m L/min;检测波长:党参炔苷267 nm,柱温:25℃;流动相:乙腈-0.05%磷酸梯度洗脱系统。结果:通过对比分析不同烘制温度下得到的炮制品,测定条件相同时,烘制党参中党参炔苷的含量高于生品;烘制温度为40℃时,蜜炙党参中的党参炔苷含量较高,烘制温度与党参中党参炔苷的含量无线性关系;蜜炙党参自然干燥品中党参炔苷含量较低。结论:不同烘制温度对党参中党参炔苷含量有一定影响;蜜炙党参最佳烘制温度为40℃。同时,采用HPLC方法测定党参中党参炔苷的含量,操作简单,结果可靠,重现性好,可作为甘肃道地药材党参及其炮制品质量控制的有效方法。
Objective: To determine the content of Lobetyolin in processed Codonopsis pilosula products under different baking temperature, and provide reliable methods for its scientific evaluation and quality control. Methods: HPLC, gradient elution and ultrasonic extraction method were used. The chromatographic conditions were as follows: the chromatographic column: Kromasil C18 chromatographic column(4.6 mm×250 mm, 5 μm), flow rate: 1.0 m L/min, detection wavelength: 267 nm,column temperature: 25 ℃, the mobile phase: acetonitrile-0.05% phosphoric acid water gradient elution system. Results:Under same conditions, the honey-processed Codonopsis pilosula products present higher Lobetyolin content than crude products, especially at the baking temperature of content of 40 ℃. There was no linearity between baking temperature and Lobetyolin content in processed Codonopsis pilosula. Conclusion: Different baking temperature can affect Lobetyolin content in processed Codonopsis pilosula. Honey-processed Codonopsis pilosula should be best to bake at 40 ℃. HPLC could be used for effectively quality control of processed Codonopsis pilosula products with simple operation, reliable results and good repeatability.
