摘要
甘油作为微生物重要的能量来源及抗逆因子,对其生存具有重要作用。gpd1基因编码甘油-3-磷酸脱氢酶(glycerol-3-phosphate dehydrogenase,G3PDH),是甘油合成代谢途径中关键的限速酶。本研究分析了gpd1基因在柑橘溃疡病菌(Xanthomonas citri subsp. citri,Xcc)致病性中的功能。对Xcc强毒菌株Xcc021和弱毒菌株Xcc049E分别进行了gpd1的缺失突变,发现gpd1缺失仅影响强毒菌株Xcc021在感病柑橘寄主上的毒性以及gpd1缺失影响Xcc在营养贫乏培养基中的生长能力,功能互补子能够恢复毒性和生长能力至野生型水平。其他毒性相关表型显示:与野生型Xcc021相比,gpd1突变体菌体的沉降能力增加,游动性降低,生物膜形成能力增强,功能互补子能够恢复这些表型至野生型水平。这表明gpd1基因在强毒菌株中是重要的毒性因子,其涉及的甘油代谢途径可能在强弱毒菌株对于寄主柑橘的毒性具有不同的作用。
As an important energy source and stress-resistance factor of micro-organisms,glycerol plays an important role in its survival.The gpd1(glycerol-3-phosphate dehydrogenase 1,gpd1)gene encodes glycerol-3-phosphate dehydrogenase(G3PDH),which is a key rate-limiting enzyme of glycerol synthesis.In this study,we analyzed the function of gpd1 gene in pathogenicity of Xanthomonas citri subsp.citri(Xcc).The gpd1 deletion mutants of Xcc hypervirulent strain Xcc021 and hypovirulent strain Xcc049E was constructed,respectively.It was found that deletion of gpd1 only affected the virulence of Xcc021 on susceptible host citrus and the growth ability of Xcc in nutrient-poor medium.The functional complementation could restore virulence and growth capacity to the wild-type levels.Some virulence relative phenotypes showed that the gpd1 mutant exhibited increased sedimentation capacity,decreased motility,and enhanced biofilm formation compared to the wild-type Xcc021,and the functional complementation were able to restore these phenotypes to the wild-type levels.This indicates that gpd1 gene is an important virulence factor in the hypervirulent strain Xcc021,and the glycerol metabolic pathway may play different roles on full virulence of the hypervirulent and hypovirulent strains on host plant citrus.
