摘要
研究组蛋白去乙酰化酶抑制剂曲古抑菌素A (TSA)对不同抗旱品种花生‘粤油-7’和‘汕优-523’根系响应干旱的影响。结果表明,与对照相比, 1μmol·L-1 TSA处理的花生根系在干旱下POD和SOD酶活力增加,抗旱相关基因AhNCED1、AhAREB1和AhDHN2的表达上升,并改变组蛋白H3ac、H3K9ac、H3K14ac的表达程度;与敏旱品种‘汕优-523’相比, TSA处理下,抗旱品种‘粤油-7’根分生区和伸长区的细胞长度增加,总根长和根表面积较大、保水能力较强、POD酶活力较高。花生2个品种的根系在响应干旱时生理反应存在的差异,可能与根内组蛋白乙酰化水平和抗旱相关基因的表达变化相关。
This paper reveals the response of root to drought stress in two peanut cultivars(‘Yueyou-7’and‘Shanyou-523’)when treated by trichostatin A(TSA),the histone deacetylase inhibitor.The results showed that,compared with the control,the activities of POD and SOD of roots in peanuts were increased under the treatment of 1μmol·L-1 TSA,then promoted the expression of drought-resistant genes,such as AhNCED1,AhAREB1 and AhDHN2,and changed the expression of histone H3 ac,H3 K9 ac,H3 K14 ac.Compared with drought-sensitive cultivar‘Shanyou-523’,the length of meristem and elongation cells in drought-resistant cultivar‘Yueyou-7’were longer when treated with TSA,and‘Yueyou-7’had longer root length and larger root surface area,stronger water retention capacity and higher POD enzyme activity.In response to drought stress,the different physiological responses of roots in different peanut cultivars may be related to the levels of histone acetylation and gene expression.
