摘要
目的:研究硫酸氢氯吡格雷与牛血清白蛋白(BSA)相互作用的荧光光谱特点,及其在硫酸氢氯吡格雷片剂含量测定中的应用。方法:应用荧光光谱法研究硫酸氢氯吡格雷与BSA的相互作用机理,建立硫酸氢氯吡格雷片剂含量测定的荧光分析方法。结果:硫酸氢氯吡格雷对BSA有荧光猝灭作用,以静态猝灭为主,两者主要靠疏水作用力相结合,结合位点数为1。硫酸氢氯吡格雷的浓度在1. 667×10-5~1. 667×10-4mol·L-1范围内与BSA的荧光强度呈良好的线性关系,相关系数为0. 998 9。用荧光光谱法测定硫酸氢氯吡格雷片剂含量,平均加样回收率为99. 57%,同一批号硫酸氢氯吡格雷片剂标示量百分含量的平均值为106. 17%。结论:采用荧光光谱法测定硫酸氢氯吡格雷片剂中主药的含量,灵敏度高,准确性好,可以作为硫酸氢氯吡格雷片剂药物含量的测定方法。
Objective:To study the fluorescence spectrometric characteristics of the interaction between clopidogrel hydrogen sulfate and bovine serum albumin(BSA)and its application in the determination of clopidogrel hydrogen sulfate tablets.Methods:Fluorescence spectroscopy was used to study the content of clopidogrel hydrogen sulfate tablets and the interaction of clopidogrel hydrogen sulfate with BSA.Results:Clopidogrel hydrogen sulfate quenched the fluorescence of BSA,with a static quenching procedure.The binding was mainly dependent on lyophobic forces and there was one binding site.The concentration of clopidogrel hydrogen sulfate showed a good linear relationship with the fluorescence intensities of BSA in a valid range from 1.667×10-5 to 1.667×10-4 mol·L-1(r=0.998 9),and the average recovery was 99.57%.The average percentage of labelled content was 106.17%in the same batch of clopidogrel bisulfate tablets.Conclusion:As a method to determine the content of the main drug in clopidogrel hydrogen sulfate tablets,fluorescence spectrophotometry is sensitive and accurate.
