摘要
目的:探讨高迁移率族蛋白B1(HMGB1)在不明原因复发性流产(URSA)患者外周血中表达水平及临床意义。方法:选取本院收治的52例URSA早孕妇女为观察组,46例正常早孕妇女为对照组。比较两组外周血辅助性T细胞17(Th17)、调节性T细胞(Treg)、Th17/Treg比值和相关指标(HMGB1、IL-17、IL-6、TGF-β及IL-10)的表达水平,分析HMGB1与各指标的相关性。结果:观察组外周血Th17细胞和Th17/Treg比值均高于对照组,Treg细胞低于对照组,差异均有统计学意义(P<0.05);观察组外周血HMGB1、IL-17、IL-6水平均高于对照组,TGF-β、IL-10水平均低于对照组,差异均有统计学意义(P<0.05);相关性分析显示,HMGB1与IL-6、IL-17、Treg、Th7均呈正相关(P<0.05),HMGB1与TGF-β、IL-10均呈负相关(P<0.05)。结论:URSA患者外周血HMGB1水平增高,其可能通过影响Th17/Treg平衡参与URSA的发病。
Objective:To investigate the expression level and its clinical significance of HMGB1 in peripheral blood of patients with unexplained recurrent spontaneous abortion(URSA).Method:A total of 52 cases of early pregnant women with URSA were selected as observation group and 46 cases of normal early pregnant women as control group.The expression levels of helper T cell 17(Th17),regulatory T cell(Treg),Th17/Treg ratio and related indicators(HMGB1,IL-17,IL-6,TGF-β and IL-10)in peripheral blood of two groups were compared,and the correlation between HMGB1 and other indicators was analyzed.Result:The level of Th17 cells and Th17/Treg ratio in peripheral blood of observation group were higher than those of control group,the Treg cells was lower than that of control group,the differences were statistically significant(P<0.05).The levels of HMGB1,IL-17 and IL-6 in peripheral blood of observation group were higher than those of control group,the levels of TGF-β and IL-10 were lower than those of control group,the differences were statistically significant(P<0.05).The correlation analysis showed that HMGB1 was positively correlated with IL-6,IL-17,Th7 and Treg cells(P<0.05),while HMGB1 was negatively correlated with TGF-β and IL-10(P<0.05).Conclusion:The increase of HMGB1 level in peripheral blood of URSA patients may be involved in the pathogenesis of URSA by affecting Th17/Treg balance.
