摘要
目的:研究在结直肠癌(CRC)组织中肿瘤坏死因子-α(TNF-α)与叉头框转录因子M1(FoxM1)的表达情况并分析其相关性。方法:选取2017年10月-2018年10月本院收治的CRC患者60例。所有患者均经手术治疗,术后均留取病灶标本(CRC组织),并取远离癌组织中心5 cm以上的组织为对照标本(癌旁组织)。以Western blot法检测CRC组织与癌旁组织TNF-α与FoxM1中的表达情况,并分析TNF-α与FoxM1在CRC组织中的相关性。结果:CRC组织TNF-α、FoxM1蛋白相对表达量均高于癌旁组织(P<0.05);组织分化程度Ⅲ、Ⅳ级和Dukes C、D期,以及有淋巴结转移、侵及浆膜层及以外的CRC组织TNF-α与FoxM1蛋白相对表达量均分别高于组织分化程度Ⅰ、Ⅱ级和Dukes A、B期,以及无淋巴结转移及未侵及浆膜层的CRC组织(P<0.05);TNF-α与FoxM1呈正相关(r=0.587,P<0.05)。结论:TNF-α、FoxM1的高表达与CRC的发生、发展有关,且两者有明显的正相关,可为CRC的诊断及预后判断提供指导,并能作为新的治疗研究靶点。
Objective:To study the expression of TNF-αand FoxM1 in colorectal cancer(CRC)and to analyze their correlation.Method:60 patients with CRC admitted to our hospital from October 2017 to October 2018 were selected.All patients were treated with surgery,and all lesions were collected postoperatively(CRC tissue),and tissues more than 5 cm away from the center of cancer tissue were taken as control specimens(adjacent tissue).The expression of TNF-αand FoxM1 in CRC tissues and adjacent tissues were detected by Western blot,and the correlation between TNF-αand FoxM1 in CRC tissues was analyzed.Result:The relative expression of TNF-αand FoxM1 protein in CRC tissues were higher than those in adjacent tissues(P<0.05).The relative expression of TNF-αand FoxM1 protein in CRC tissues with differentiation degreeⅢ,Ⅳlevel,Dukes C,D period,lymph node metastasis,invasion into serous layer and beyond were higher than those in CRC tissues with differentiation degreeⅠ,Ⅱlevel,Dukes A,B period,no lymph node metastasis and no serous layer invasion,respectively(P<0.05).TNF-αwas positively correlated with FoxM1(r=0.587,P<0.05).Conclusion:The high expression of TNF-αand FoxM1 is related to the occurrence and development of CRC,and they are positively correlated.They can provide guidance for the diagnosis and prognosis of CRC,and can be used as new therapeutic research targets.
