摘要
目的:探究电针预处理对动脉粥样硬化型兔腹腔巨噬细胞受体CD36及SR-A1的基因和蛋白表达的影响。方法:将32只雄性新西兰兔随机分为3组,即空白对照组(7只)、造模组(19只)、电针预处理组(6只)。兔动脉粥样硬化的模型通过高脂饲料喂养加行颈总动脉球囊损伤术。造模组再随机分为模型对照组、阳性药物组、电针治疗组,每组6只。造模组经8周造模成功后,进行4周的干预治疗。电针预处理组适应性喂养1周后,进行电针预处理,针刺双侧的内关穴、足三里穴以及关元穴,双侧内关穴、足三里穴连接电针仪20 min,采用疏密波(4 Hz/20 Hz,1 m A),每日1次,针刺6 d,休息1日,针刺3周后,再经8周造模,造模成功后,再给与电针治疗组相同的干预治疗。采用免疫组化法检测各组兔腹腔巨噬细胞受体CD36及SR-A1的蛋白表达情况;采用RT-PCR检测各组兔腹腔巨噬细胞受体CD36及SR-A1的基因表达情况。结果:与正常组比较:模型组的腹腔巨噬细胞受体CD36及SR-A1的基因和蛋白的表达均明显升高(P<0.05);与模型组比较:电针治疗组、阳性药物组的腹腔巨噬细胞受体CD36及SR-A1的基因和蛋白的表达均明显降低(P<0.05),电针治疗组与阳性药物组比较差异无统计学意义(均P>0.05);与电针治疗组、阳性药物组相比:电针预处理组的腹腔巨噬细胞受体CD36及SR-A1的基因和蛋白的表达均明显降低(P<0.05)。结论:电针预处理组能明显降低动脉粥样硬化兔腹腔巨噬细胞受体CD36及SR-A1的表达,这可能是针刺预防动脉粥样硬化疾病的机制之一。
Objective:This study has explored the effect of electroacupuncture preconditioning on gene and protein expression of atherosclerotic rabbit peritoneal macrophage receptor CD36 and SR-A1.Methods:Thirty-two male New Zealand rabbits were randomly divided into 3 groups:blank control group(7),modeling group(19),electroacupuncture pretreatment group(6).Atherosclerosis in rabbits was treated with high-fat diet and carotid artery balloon injury.The model group was further randomly divided into subgroups containing a model control group,a positive drug group,and an electroacupuncture treatment group with 6 rabbits in each group.The model group was given intervention treatment for 4 weeks after successful modeling for 8 weeks.After one week of adaptive feeding,the electro-acupuncture pretreatment group was treated with electro acupuncture,specifically.The acupuncture was given at Neiguan(PC6),Zusanli(ST36)and Guanyuan(RN4)respectively,bilateral Neiguan(PC6)and Zusanli(ST36),of which were connected with electro-acupuncture apparatus for 20 minutes.The density wave(4 Hz/20 Hz,1 m A)was implemented at a treatment by once a day,6 days continuous acupuncture,and 1 day rest.After 3 weeks acupuncture,a subsequent 8–week model building was tested.Finally,the same intervention treatment as the electroacupuncture treatment group was given after successful modeling.The protein expressions of CD36 and SR-A1 in peritoneal macrophages of rabbits in each group were detected by immunohistochemical method,and the gene expressions of CD36 and SR-A1 in peritoneal macrophages of rabbits in each group were detected by RT-PCR.Results:The mRNA and protein expression of peritoneal macrophage receptors CD36 and SR-A1 in the model group increased significantly(P<0.05)in comparison with the normal group.Additionally,the mRNA and protein expression of peritoneal macrophage receptors CD36 and SR-A1 in the electroacupuncture group and the positive drug group were significantly decreased(P<0.05)compared with the model group and the difference in the comparison between the electroacupuncture treatment group and the positive drug group was not statistically significant(all P>0.05).At last,compared with the electroacupuncture group and the positive group,the mRNA and protein expression of the peritoneal macrophage receptor CD36 and SR-A1 in the electroacupuncture preconditioning group were significantly decreased(P<0.05).Conclusion:Electroacupuncture preconditioning can significantly reduce the expression of peritoneal macrophage receptors CD36 and SR-A1 in atherosclerotic rabbits,which may be one of the mechanisms of acupuncture to prevent atherosclerotic diseases.
