大黄附子汤作用机制的靶标与通路探讨 被引量:9

摘要 目的:通过系统药理学方法探讨大黄附子汤的作用机制及原理。方法:通过中药系统药理学分析平台(TCMSP)数据库提取大黄附子汤中的活性成分和有关的靶标蛋白,通过运用软件Cytoscape 3. 5. 1构筑大黄附子汤有效成分与靶标关系网络进行拓扑学分析,使用STRING数据库进行蛋白之间相互作用(PPI)网络的构筑与分析,借助David数据库进行KEGG通路功能富集分析。结果:大黄附子汤的核心化合物有山奈酚(kaempferol)等,重要的靶标有前列腺素G/H合成酶2(Prostaglandin G/H synthase2)等,大黄附子汤有效成分中关键蛋白主要有JUN、RELA、TP53、CYP1A1、MAPK1等,KEGG结果表明大黄附子汤主要作用于化学致癌、细胞色素P450及视黄醇代谢等相关信号通路。结论:大黄附子汤主要通过多种途径、多种信号通路作用于多种靶点发挥抗炎、抗癌及治疗功能性便秘等作用。 Objective:To investigate the mechanism and the principle of Dahuang Fuzi Decoction by systemic pharmacological method.Methods:The active ingredients and related target proteins in Dahuang Fuzi Decoction were screened by the database of pharmacology analysis platform of Chinese medicine system(TCMSP).The network visualization software Cytoscape 3.5.1 was used to construct the topological analysis of the effective component-target interaction network of Dahuang Fuzi Decoction.The construction and analysis of protein and protein interaction(PPI)network are carried out with the STRING online database.The function enrichment analysis of KEGG pathway was carried out with the David database.Results:The results showed that core compound of Dahuang Fuzi Decoction have kaempferol,beta sitosterol;the key target is prostaglandin G/H synthase 2,heat shock protein 90.The key proteins in the effective components of Dahuang Fuzi Decoction are JUN,RELA,TP53,CYP1 A1,MAPK1 and so on.The results of KEGG show that Dahuang Fuzi Decoction is mainly involved in chemical carcinogenesis,cytochrome P450 and retinol metabolism and other related signaling pathways.Conclusion:Dahuang Fuzi Decoction mainly through a variety of ways,a variety of signaling pathways to play a variety of targets to play anti-inflammatory,anti-cancer and therapeutic functional constipation.
