摘要
目的:探讨温胆汤含药血清对谷氨酸环境下星型胶质细胞凋亡及NRG1、ErbB4表达的影响。方法:SD大鼠灌胃相应药物或生理盐水后制备含药血清,灭活,过滤除菌,EP管(4mL)分装备用。以正常组、模型组、氯氮平20mg/kg含药血清、温胆汤40g/kg、20g/kg、10g/kg含药血清培养,干预谷氨酸环境下星形胶质细胞,流式细胞仪检测星形胶质细胞凋亡;Western bloting、Real-time PCR分别测定星型胶质细胞NRG1、ErbB4蛋白、磷酸化和mRNA表达。结果:温胆汤各浓度含药血清组均能明显减少星形胶质细胞凋亡;降低NRG1、ErbB4蛋白和mRNA表达,提高其磷酸化表达。结论:温胆汤10%含药血清可有效调节NRG1、ErbB4表达,从而调控NRG1-ErbB4信号通路的功能,保护神经细胞,这也许是温胆汤能够治疗精神分裂症的作用机制之一。
Objective:To investigate the effect of Wendan Decoction-containing serum on astrocyte apoptosis and expressions of NRG1 and ErbB4 in glutamate environment.Methods:SD rats were administered with corresponding drugs or saline.After the administration,serum containing drug was centrifuged,inactivated,filtered,sterilized and collected in EP tubes(4 mL).Astrocytes were cultivated in six different groups:the normal group,the model group,20 mg/kg clozapine group,40 g/kg、20 g/kg and 10 g/kg Wendan Decoction groups.The apoptosis of astrocytes was detected by flow cytometry.The expressions of NRG1,ErbB4 protein,phosphorylation and mRNA in astrocytes were detected by Western blotting and Real-time PCR,respectively.Results:All doses of serum containing Wendan Decoction significantly reduced the apoptosis of astrocyte,decreased the expressions of NRG1,ErbB4 protein and mRNA,and increased the phosphorylation expressions of NRG1 and ErbB4.Conclusion:10%serum containing Wendan Decoction can effectively regulate the expressions of NRG1 and ErbB4,thereby regulate the function of NRG1-ErbB4 signal pathway and protect nerve cells,which may be one of the mechanisms of Wendan Decoction in treating schizophrenia.
