摘要
目的 观察中药制剂舒安注射液 (SAI)对脑出血的救治作用及其对脑出血后脑内神经生长因子 (NGF)和白细胞介素 - 1β(IL - 1β)表达的影响。方法 采用大鼠自体股动脉血微量注射入内囊造成脑出血模型 ,SAI给药治疗 7d,记录脑出血模型大鼠的神经病学评分 ;通过 Northern杂交与 ELISA方法检测出血后脑内 NGF与 IL - 1βm RNA及其蛋白的表达与含量。结果 SAI治疗后 ,大鼠神经病学评分明显低于治疗前及生理盐水组 ;大鼠脑组织含水量 ,Na+含量及脑指数均显著低于生理盐水对照组。大鼠脑出血后第 1天 NGF与 IL - 1β m RNA表达增加 ,第 2天 NGF、IL - 1βm RNA下降至正常组水平 ,第 4天 ,7天低于正常组水平 ;IL - 1β m RNA下降缓慢 ,至第 7天始降至正常组水平。 NGF和 IL - 1β蛋白含量在出血后第 1天显著增高 ,至第 7天仍高于正常水平。 SAI治疗组 NGF m RNA与蛋白含量变化同模型组 ,IL - 1β m RNA和蛋白在出血后各时间点均低于模型组。结论 SAI可明显促进脑出血大鼠的神经功能的恢复 ,其作用机制可能与抑制 IL- 1β表达和上调 NGF表达有关。
Objective To explore the effects of SHU AN Injection (SAI) on experimental intracerebral hemorrhage (ICH) and the expressions of nerve growth factor (NGF) and interneukin-1β (IL-1β) after ICH injury in rat.Methods Model of capsuloganglionic hemorrhage in rats was made through injection of 120 μl auto-femoral arterial blood, and the rats with ICH were treated with SAI and nimodipine injection for 7 days. Northern blot and enzyme linked immunosorbent assay (ELISA) were employed to detect the mRNA and proteins of NGF and IL-1β in rat brain tissues.Results After the SAI treatment, the neurologic grades and the concentrate of water and Na + in cerebral tissue were lower significantly compared with control group. The expressions of NGF and IL-1β mRNA increased on the first day after ICH injury, decreased to the normal level on the second day and decreased to the level under normal on the 4th and 7th . Both NGF protein and IL-1β protein contents increased markedly from 1 day after hemorrhage, still higher than normal 7 days later. The changes of NGF mRNA and protein content of SAI group were same as model group, but the IL-1β mRNA and protein in SAI group were significantly lower than those in model group at all time points. Conclusions SAI could promote the neural function recovery following ICH in rats significantly, and its mechanism probably was related to repress IL-1βmRNA expression and upregulate the expression of NCF.
