摘要 目的 :探讨OxLDL在动脉粥样硬化血管局部免疫病理损伤中的作用机制。方法 :采用地高辛标记的寡核苷酸探针原位杂交技术 ,观察OxLDL对人外周血单核细胞MCP 1、PDGF、bFGF、IL 10 4种致炎细胞因子mRNA表达的影响。结果 :OxLDL(10~ 30mg L)同人外周血单核细胞孵育 2 4小时后能够以剂量依赖性方式显著提高MCP 1、bFGF、PDGF、IL 10 4种致炎细胞因子mRNA表达 (均P <0 0 0 1)。结论 :OxLDL能以剂量依赖性方式直接刺激人外周血单核细胞分泌致炎细胞因子mRNA。 Objective:To investigate the function of oxidized low density lipoprotein(OxLDL) in the formation of atherosclerotic immuno-pathological lesions in the local blood vessel.Methods:With the Digoxenin-labeled Oligonucleotide-probes In Situ Hybridization,studied the effects of OxLDL on the mRNA expression of proinflammatory cytokines including MCP-1, bFGF, PDGF and IL-10 in the human peripheral blood monocyte. Results:Monocyte was significantly increased the mRNA expression of MCP-1, bFGF, PDGF and IL-10 in a dose-dependent manner after incubating with OxLDL(10~30 mg/L)for 24 hours(P<0.001).Conclusion:OxLDL can directly stimulate human peripheral blood monocytes to excrete proinflammatory cytokines mRNA in a dose-dependent manner.
