摘要 目的 :建立人瘦素的放射免疫分析 (RIA)方法并用于临床。 方法 :以重组人瘦素 (LPT)小剂量多次刺激新西兰白兔制备抗血清 ,以间接碘标记法制备12 5I LPT ,然后建立LPT的RIA法并用于临床。 结果 :LPT的抗血清工作液稀释度为 1∶6 4 0 0 ;12 5I LPT的放射性比活度为 1.4 3MBq/ μg ,12 5I的总标记率为 37.8% ;LPT的RIA各项技术指标均符合要求 ;临床检测结果 ,正常男性为 (4 .9± 2 .7) μg/L ,女性为 (8.9± 4 .0 ) μg/L ,性别差异显著 (P <0 .0 1)。 结论 :LPT的RIA方法各项考核指标均较好 。 Objective: To study the radioimmunoassay(RIA) of leptin(LPT) in clinical use. Methods: Anti-LPT serum was produced by repeated stimulation of small dose rhLPT to New Zealand rabbits. 125I-LPT was iodinated with BH reagent by indirect iodination labelling,and then RIA of LPT was established in clinics. Results: The working dilution of anti-LPT serum was 1 ∶ 6 400; the specific radioactivity of 125I-LPT was 1.43 MBq/μg, the total labeling rate of 125I was 37.8%; the technical specifications of RIA of LPT could satisfy the needs of RIA; the assay values of normal male and female was (4.9±2.7)μg/L and(8.9±4.0)μg/L respectively with marked sex difference (P<0.01). Conclusion:The test specifications of RIA of LPT could satisfy the clinical needs for reducing the harm of direct iodination to LPT.
