摘要
目的 探讨中国南方汉族人基质金属蛋白酶-9(MMP-9)基因多态性与慢性阻塞性肺疾病(COPD)易感性的关系。方法 应用限制性片段长度多态性(RFLP)技术,检测MMP-9启动子基因型在100例COPD患者和98例健康吸烟者中的频率。结果 COPD中同源野生型(C/C)、杂合型(C/T)频率分别是86%、14%,健康人的频率分别是98%、2%,两组基因频率分布差异具有显著性(P<0.01)。等位基因C频率分别是93%比99%,等位基因T频率分别是7%比1%,两组等位基因的分布差异也具有显著性(P<0.05)。结论 MMP-9启动子-1562位的多态性可能与中国南方汉族人群COPD易感性相关。
Objective To investigate the correlation between matrix metalloproteinase-9 ( MMP-9) gene polymorphism and chronic obstructive pulmonary disease (COPD) susceptibility in Chinese population of Han nationality of southern China. Methods Examine the frequency of the genetype of the promoter of MMP-9 (-1562C/T) in 100 COPD patients and 98 smoking control subjects without COPD by restriction fragment length polymorphism. Results The frequencies of polymorphic genotypes in promoter of MMP-9 in the COPD group were C/C 86% and C/T 14% , and the frequencies of polymorphic genotypes in control group were C/C 98% and C/T 2% , There were significant differences between the two groups(P <0. 01) . The differences in allele frequencies were also significant between the two groups (C allele frequency : 93% vs 99% , T allele frequency: 7% vs 1% , P < 0. 05). Conclusion The genetic polymorphism in promoter of MMP-9 gene was associated with susceptibility to COPD in the Chinese population of Han nationality of southern China.
