摘要
目的 :研究鸡肝散总黄酮 (TotalflavoneofElsholtziablanda ,TFEB)抗大鼠实验性心律失常的作用。方法 :大鼠采用结扎冠脉左前降支 (leftanteriordescendingcoronaryartery ,LAD)、iv垂体后叶素 (pituitrinum ,pit)、恒速iv乌头碱诱发大鼠心律失常的动物模型 ,记录Ⅱ导ECG ,分别观察大鼠心律失常发生的情况。结果 :TFEB 15 0、2 0 0mg/kg能显著减少结扎后 15min内室性早博(ventricularextrasystole ,VE)的发生次数 ,减少发生室性心动过速 (ventriculartachycardia ,VT)的动物数 ,降低ivpit所致心律失常发生的百分率 ;TFEB各剂量组均能极显著提高乌头碱致大鼠VE的阈值 ,TFEB 3 0 0mg/kg还能显著提高乌头碱诱发大鼠VT、VF和心脏停搏 (heartstop ,HS)的阈值。 结论 :TFEB具有抗实验性心律失常作用。
Object: To study the effect of total flavone of Elsholtzia blanda (TFEB) on experimental arrhythmia in rats. Methods: Male rats were given TFEB by ig for 5d, various arrhythmia models were induced by left anterior descending coronary artery(LAD) ligation, iv pituitrinum(pit), or iv aconitine in a constant speed of 2 μg/min. LeadⅡdynamic ECG was recorded and arrhythmia was observed accordingly to assess the effect of TFEB. Results: At the dosage of 150, 200mg/kg, TFEB could markedly reduce the incidence of ventricular extrasystole(VE) and ventricular tachycardia(VT) within 15min after LAD ligation(P<0.05~0.01),at the same time ,the incidence of ventricular fibrillation (VF) also showed a reduced tendency, although there was no marked difference compared with control group(P>0.05). Compared with the high arrhythmia frequency of 82.4% in the control group, a markedly lower rate of arrhythmia in the TFEB groups, especially at the dosage of 200 mg/kg(P<0.05)after iv pit were observed. The threshold dosage of aconitine which lead rats VE was significantly increased by TFEB in dose-dependent manner(P <0.01). Besides, at the dosage of 300 mg/kg TFEB could also markedly increase the theshold value which induced VE, VT and heart stop(HS)(P<0.05~0.01). Conclusion: TFEB could not only prevent ischemic arrhythmia induced by LAD ligation and iv pit, but also increased the threshold value of arrhythmia induced by aconitine. TFEB had the effect of anti-arrhythmia.
