摘要
从操作耗时、所得RNA产量和质量等方面比较了5种RNA提取方法提取甜橙叶片、幼果、成熟果实果皮和汁囊组织RNA的效果。结果表明,改良Bugos法能从上述4种组织中获得较高产量和质量的RNA。用该方法提取的温州蜜柑成熟果实果皮RNA经RT-PCR扩增成功得到了蔗糖磷酸合成酶cDNA片段。该方法还可适用于猕猴桃、梨、甜瓜、龙眼、苹果和桃果肉RNA的提取。
methods f or RNA extraction from4different kinds of Citrus sinensis tissues,including f ruitlet,leaf,peel and juice sacs of ripe fruit,were evaluated upon time consu mption,yield and quality of RNA isolated.A modified Bugos method was found to be the best one among the5methods evaluated.Only with the modified Bugos metho d,higher amount and quality of RNA could be successfully extracted from all4k inds of tissues tested.The RNA obtained from peel of ripe Citrus unshiu fruit w ith the modified Bugos method was successfully used to amplify a cDNA fragment of sucrose phosphate synthase by RT-PCR.The method was found to be also suitab le for extracting RNA from pulp of ripe kiwifruit,pear,melon,longan,apple an d peach.
