摘要
利用细胞悬浮培养和TTC分析法,进行葡萄细胞抗寒性筛选的研究结果表明,3个供试品种,未经低温筛选的细胞低温致死温度为-3℃左右。经过-3至-7℃人工低温分级筛选,可以筛选出低温致死温度在-5、-6和-7℃左右的抗寒细胞系。在-3至-7℃范围内,筛选温度越低,所获抗寒细胞系的抗寒性越强,其低温致死温度也越低。当筛选温度下降至-8℃时,3个供试品种的细胞残存量都极微,没有筛选出抗寒细胞系。
An increasing number of mutant cell lines of plants have been selected by in vitro for resistance to herbicide.,disease and salt tolerance. But few reports had been published in the area of cold resistant screening. The result of this investigation will open up a way for the selection of new varieties of crops to tolerance to cold stress through cell suspension culture.
The effective media components, noncontaminated seeding procedure and the method of TTC assay for cold-resistant test of cells were successfully used in this investigation. The cell cold-killing temperatures of the three grape varieties tested in this investigation were around -3℃
Cells to be cold screened in subszero temperatures, from -3℃ to -7℃, could effectively select the mutant cold-resistant cell lines with the cold-killing temperatures of - 5, - 6 and - 7℃. The lower the screening temperature, the higher cold-resistance of the mutant cell line obtained. At the temperature of - 8℃ or lower,few cells could survived.
