摘要
目的 :观察顺铂对HeLa细胞端粒酶活性、凋亡及其p5 3、bcl 2表达的影响 ,以揭示顺铂抗宫颈癌的机理。方法 :运用体外细胞培养技术 ,以不同浓度的顺铂作用于培养的HeLa细胞 72h ,通过端粒酶重复序列扩增 -酶联免疫吸附法 (TRAP ELISA)检测细胞端粒酶活性 ,以及流式细胞术观察细胞凋亡及p5 3、bcl 2的表达。结果 :顺铂能以浓度依赖方式下调端粒酶活性并诱导细胞凋亡 ,使其bcl 2基因的表达显著降低 ,而p5 3基因的表达则增强。端粒酶活性下调与细胞凋亡及其p5 3、bcl 2基因表达紧密相关 ,P <0 0 1。结论 :顺铂能下调端粒酶活性 ,诱导细胞凋亡 ,使bcl 2表达降低和p5 3表达增强 ,这可能是顺铂抗癌作用的重要机制之一。
Objective To investigate the effect of cisplatin on telomerase activity, apoptosis and p53,bcl-2 gene expressions of cervical cancer HeLa cells in order to find out the mechanism of its anticancer action.Methods By techniques of cell culture in vitro,HeLa cells were treated by cisplatin in different concentration for 72 hours,then the cell telomerase activity was measured by Telomeric Repeat Amplification Protocol with ELISA(TRAP-ELISA) and the apoptosis,p53 and bcl-2 gene expressions of the cell were observed by Flow-Cytometry (FCM) respectively.Results Cisplatin down-regulated the telomerase activity of HeLa cells,induced the apoptosis of the cell by concentration-dependent fashion,obviously decreased the expression of bcl-2 gene protein and increased the expression of p53 gene protein.There was a good correlation between the telomerase activity and the apoptosis and the expressions of p53 and bcl-2 gene protein.Conclusions Cisplatin could down-regulate the telomerase activity,induce the apoptosis,decrease the expression of bcl-2 gene protein and increase the expression of p53 gene protein,which may be one of important mechanisms of cisplatin’s anticancer action.
