摘要
目的 研究可生物降解聚合物 PEL A作为控制释放的基因载体。方法 合成聚乳酸 -聚乙二醇二元共聚物(PEL A) ,包裹质粒 p CH110 ,探讨了 PEL A作为基因载体包裹 DNA的各种影响因素 ,这种控制释放体系的体外降解及释放行为以及对其细胞毒性及体外转染效率进行了测定。结果 制备的 PEL A微球平均粒径 1μm~ 3μm,包裹效率为6 2 %。采用包裹了质粒 p CH110的微球对 COS- 1细胞的细胞毒性及转染实验中得出 :PEL A的细胞毒性较小 ,转染效率较高 ,且能持续表达 96 h,缓释效果较明显。结论 可生物降解聚合物材料 PEL
Objective To study the biodegradable polymers PELA as gene controlled released coatings for gene transfer. Methods The poly(Dl lactic acid) co poly(ethylene glycol) (PELA) was synthesized and prepared as the coating of plasmid pCH110 in the transfection. All kinds of factors affecting the loading efficiency, cytotoxicity, transfection efficiency and the course of the degradation and release in vitro were discussed. Results The average diameter of microspheres of PELA was 1~3μm. The loading efficiency was 62%. The transfection efficiency of the pCH110 delivery system for COS 1 cells was high and had few cytotoxicity. After transfection, the X gal assay was positive for 96 h. Conclusion The biodegradable polymeric materials PELA as gene carriers possess the potential superiority.
