HLA-DQA1基因多态性与支气管哮喘及慢性支气管炎易感性的比较研究 被引量:8

摘要 目的 研究支气管哮喘与慢性支气管炎患者HLA DQA1等位基因的异同。方法 采用限制性片断长度多态性聚合酶链反应技术 (PCR RFLP) ,对 4 0例支气管哮喘及 2 1例慢性支气管炎江西籍汉族患者进行HLA DQA1等位基因频率的检测 ,与 4 2例江西籍汉族正常人进行比较分析。结果 与正常对照组比较 ,支气管哮喘及慢性支气管炎组HLA DQA1 0 4 0 1基因频率均明显增高 (P <0 0 1) ,且在支气管哮喘组中 ,HLA DQA1 0 6 0 1基因频率明显增高 (P <0 0 1) ;支气管哮喘组与慢性支气管炎组比较 ,HLA DQA1 0 4 0 1、 0 6 0 1基因频率的分布无显著性差异 (P >0 0 5 )。其余各等位基因频率在三组间分布也无显著性差异 (P >0 0 5 )。结论 江西汉族人群的HLA DQA1 0 4 0 1、 0 6 0 1基因可能为支气管哮喘的易感基因 ,HLA DQA1 0 4 0 1基因可能为慢性支气管炎的易感基因 ,提示哮喘和慢性支气管炎有部分相同的免疫遗传机制。 Objective To investigate the role of HLA-DQA1 allele in susceptibility to asthma and chronic bronchitis.Methods 40 patients with asthma and 21 patients with chronic bronchitis from the Han people in Jiangxi province were investigated.HLA-DQA1 alleles were determined using PCR-RFLP genotype method and the patients' data were compared with that of health controlled group. Results Compared with the health controls,the frequencies of HLA-DQA1 *0401 alleles were significantly increased in asthma group (P<0.01) and chronic bronchitis group (P<0.01) and the frequency of HLA-DQA1 *0601 allele was significantly increased in asthma group (P<0.01).However there were no significant differences in the frequencies of HLA-DQA1 *0401 *0601 alleles in asthma group compared with chronic bronchitis group (P>0.05).The frequencies of other DQA1 alleles were not significantly different in these groups.Conclusions Our data indicate that HLA-DQA1 *0401, *0601 alleles may be susceptible genes to asthma while HLA-DQA1 *0401 allele may be susceptible gene to chronic bronchitis among the Hans in Jiangxi provine,suggesting common genetic background in asthma and chronic bronchitis.
