人胰岛素基因重组杆状病毒在家蝇中表达的研究 被引量:1

摘要 目的 :利用杆状病毒表达系统进行人胰岛素基因在家蝇中表达的研究。方法 :用携带有人胰岛素基因的重组杆状病毒感染家蝇幼虫 ,用放射免疫技术、Tricine SDS PAGE和Western blot技术对蝇蛆胰岛素表达水平进行检测。结果 :重组的杆状病毒感染家蝇幼虫后 ,蝇蛆中有人胰岛素的表达 ,表达量为 37.4 0 1μIU/ml,占蝇蛆总蛋白的 0 .5 83%。结论 :利用杆状病毒表达系统可以在家蝇蝇蛆中表达人胰岛素。 Objective: To study the expression of human insulin gene with baculovirus expression system in housefly. Methods: The recombinant baculovirus contained human insulin gene were used to infect housefly larva. The human insulin was detected by using Tricine-SDS-PAGE and Western-blot. Results: Human insulin gene expressed in infected housefly larva. The most expression level was 37.401 μIU/ml. Conclusion: Human insulin gene could express in housefly larva with baculovirous expression system.
