摘要
背景:中医学认为针刺涌泉穴(K11)可以补肾活血,从而延缓衰老,但其疗效的分子机制尚不清楚。 目的:研究针刺对衰老造模大鼠细胞凋亡相关基因p53,bcl-2表达的影响,探讨针刺涌泉穴延缓衰老的分子机制。 设计:随机对照实验研究。 地点、材料和干预:本实验在四川大学生命科学学院分子生物学实验室完成。本实验选用雄性5周龄SD大鼠16只。将大鼠按随机抽签法分为正常对照组、衰老模型组、针刺治疗组与针刺对照组,每组4只。衰老模型组、针刺治疗组与针刺对照组动物均于每日皮下注射100g/LD-半乳糖0.5mL,连续注射7周。针刺治疗组在注射D-半乳糖的同时每周1至周5电针涌泉穴,频率10Hz,强度2mA,3min/次。针刺对照组的处理除电针涌泉穴旁的非穴位区以外,其他处理同针刺治疗组。正常对照组除每天注射生理盐水外,进行正常饲养。 主要观察指标:电针涌泉穴与非涌泉穴旁非穴位区对衰老大鼠p53和bcl-2基因表达的影响。通过p53和bcl-2的反转录-聚合酶链式反应(reverse transcription and polymerase chain reaction,RT-PCR)分析来表现上述影响。 结果:电针能明显改善患鼠症状;D-半乳糖造模引起p53基因表达上调,经电针涌泉穴后又可使其下调;bcl-2基因的表达不受D-半乳糖造模和电针的影响。 结论:针刺涌泉穴对衰老大鼠的p53基?
BACKGROUND:Traditional Chinese medicine considers acupuncture of yongquan(KI1) acupoint has the effect of delay aging by benefiting kidney and invigorating the circulation of blood,but the mechanism of the effect is unclear.OBJECTIVE:To discover the mechanism of delaying aging by acupuncture of yongquan acupoint,the effect of electroacupuncture of yongquan acupoint on the expression of p53 and bcl-2 in the aging model rats was researched.DESIGN:Randomized control experimental research.SETTING,PARTICIPANTS and INTERVENTION:The present study was carried out in the Laboratory of Molecular Biology,College of Life Sciences,Sichuan University.
A total of 16 males,5 weeks old SD were obtained.The rats were divided into normal control group,aging model group,electroacupuncture group,and acupuncture control group by random sortition method.Every group had 4 rats respectively.The rats of aging model group,electroacupuncture group,electroacupuncture control group were given 0.5 mL 100 g/L D-galactose daily subcutaneous injection for 7 weeks.After subcutaneous injection of 0.5 mL 100 g/L D-galactose,the electroacupuncture group was electroacupunctured at yongquan acupoint with a frequency of 10 Hz,and 2 mA current for 3 min once a day from Monday to Friday every week.The rats of the electroacupuncture control group were treated as the same that of the electroacupuncture group nothing but electroacupuncture on the non-point nearly besides the yongquan acupoint.The rats in the normal control group were fed normally nothing but receiving 0.5 mL saline daily subcutaneous injection for 7 weeks.MAIN OUTCOME MEASURES:The effect of electroacupuncture of yongquan acupoint on the expression of p53 and bcl-2 was studied.Reverse transcription polymerase chain reaction(RT-PCR) was used to monitor the influence.
RESULTS:The symptoms of the aging model rats could be relieved obviously by electroacupuncture.p53 was up regulated in the D-galactose aging model rats' liver and partly down regulated in the liver of the rats treated with electroacupuncture.bcl-2 expression was not influenced by D-galactose-model-making and electroacupuncture.
CONCLUSION:The p53 gene expression in aging model rats is suppressed by electroacupuncture of yongquan acupoint
