克山病病区粮饲养大鼠心肌收缩蛋白改变的分子机制研究 被引量:2

摘要 从克山病病区粮和非病区粮饲养大鼠心肌中提取总 RNA,分析其中的α和β—CMHCmRNA 含量的变化。结果发现,与非病区粮组相比,病区粮组大鼠心肌中的α—CMHC m-RNA 含量逐渐降低,而β—CMHC mRNA 含量逐渐升高。在大鼠心肌 CM—ATP 酶活性的分析中发现,病区粮组大鼠 CM—ATP 酶活性的降低与 CM 基因表达变化之间具有良好的相关性。从而证明 CM—ATP 酶活性的变化是由于α—CMHC 基因表达减少和β—CMHC 基因表达增多,进而导致 CM 同工酶由 V_1向 V_3转化的结果。 To extract total RNA from myocardium of the rats fed on grains both fromKD area and Non-KD area,the dynamic changes of a and β-CMHC mRNAcontent was analyzed in rat myocardium.The result showed that,comparedwith the rats of Non-KD group,α-CMHC mRNA content reduced,and β-CMHC mRNA content increased (P<0.05) in the myocardium of the rats fedon KD area grains.It was found that in rats fed on KD area grains,thedynamic comparison of CM-ATPase activities with CM-gene expression sug-gested that the two changes had excellent relationship.The results indicatedthat the decrease of myocardial CM-ATPase activity in KD was due to thedecrease of a-CMHC gene expression and the increase of β-CMHC gene expres-sion which further induced the transformation of CM isoemzymes from V_1 to V_3.
