摘要 目的 :探讨 PTEN 对人脑胶质瘤血管生成和肿瘤生长的抑制作用。 方法 :用含 PTEN基因的重组腺病毒载体Ad PTEN,体外转染人胶质瘤细胞 U87,Western印迹分析检测其表达 ,采用鸡胚绒毛尿囊膜实验检测其对血管生成的作用 ,体内实验检测腺病毒对肿瘤体积的影响 ,同时以免疫组化法检测 因子相关抗原和基质金属蛋白酶 (MMP- 2 )的表达 ,并与U87组 (不予任何处理 )和 Ad X- gal转染组相比较。结果 :U87转染 Ad PTEN腺病毒后 PTEN表达上调 ,U87组的血管指数、体内肿瘤体积、 因子相关抗原和 MMP- 2的表达病理指数均显著多于 Ad X- gal和 Ad PTEN组 (P<0 .0 5 ,P<0 .0 1 )。结论 :PTEN可抑制肿瘤血管生成 ,并抑制体内肿瘤的生长。提示 Objective: To evaluate the effects of adenovirus mediated PTEN on the angiogenesis and growth of malignant glioma cells U87. Methods: PTEN expression was determined by Western blot after it was used to infect human U87 glioma cells with the adenovirus vector in vitro . Chick chorioallantoic membrane (CAM) test was used to testify the effect of PTEN on angiogenesis of tumor cells. The U87 cells infected with adenovirus was injected into nude mice brain to observe the tumor growth.The expression of factor Ⅷ related antigen and MMP 2 was examined by immunohistochemistry. Results: PTEN expression increased stably in PTEN transfected glioma cell lines.Compared with AdX gal group and AdPTEN group,the vessel index,tumor volume and the expression of factor Ⅷ related antigen and MMP 2 in pure U87 group were significantly increased( P <0.05, P <0.01). Conclusion: AdPTEN can effectively inhibit the angiogenesis of glioma and the growth of tumor, suggesting PTEN may be useful for the treatment of malignant glioma through anti angiogenesis.
