摘要 目的 :探讨成熟卵泡液、促性腺激素对人类生殖泡期 (germinalvesicle,GV)不成熟卵母细胞体外成熟 (invitromaturation ,IVM)的影响。方法 :在基本培养液中加入 5 0 %成熟卵泡液 (folliclefluids ,FF) (Ⅰ组 )、或促性腺激素 (gonadotrophin ,Gn) (Ⅱ组 )或单用基本培养液即对照组 (Ⅲ组 )用于GV期卵母细胞的体外培养 ,观察成熟率及进一步发育能力。结果 :三组之间GVBD(生殖泡期破裂 )率差异无显著性 (P >0 .0 5 ) ,MⅡ期率和受精率 ,Ⅰ、Ⅱ组明显高于Ⅲ组 (P <0 .0 1)、Ⅰ组与Ⅱ组之间无差异 (P >0 .0 5 ) ;Ⅰ组卵裂至 8细胞的比例为 6 2 .5 % ,明显高于Ⅱ组的33.3% (P <0 .0 1)。结论 :GV期卵母细胞尽管在基本培养液中可完成核成熟 ,但胞质不成熟 ,加入 5 0 %FF或Gn均能促卵母细胞的成熟 ,两者相比 5 0 %FF效果更好。 Objective:To study the effects of FF on the human immature oocyte. Methods:GV stage oocyte-cumulus complexes(OCCs) were retrieved from pregnant women undergoing cesarean section,randomly divided into three groups group Ⅰ(n=34):M199+50%FF,groupⅡ(n=34):M199+0.075 iu/ml HMG+0.5 iu/ml HCG+17βE 2+15%FCS,and groupⅢ(n=35):M199+15%FCS as control.The OCCs were evaluated for meiotic maturation using an inverted microscope. Oocytes were observed at 24 hours and repeated at 48-52 hours after culture.The germinal vesicle breakdown (GVBD)stage and MⅡstage were recorded. MⅡstage oocytes were fertilized with standard method,observed for fertilization and cleavage every 24 hours for three days. Results: GVBD rate showed no statistically significant differense between three groups. MⅡstage rate and fertilization rate in groupⅠ,Ⅱwere increased significantly than groupⅢ ( P <0.01),8-cell embryo cleavage rate was much higher in groupⅠthan in groupⅡ.Conclusion:the mature FF seems to increase in vitro maturation rate of immature human oocytes,and can improve oocytes developmental competence.
