摘要 丙型肝炎病毒 (HCV)是引起病毒性非甲非乙肝炎的主要病原 .全世界约有 1.7亿HCV的感染者 ,我国估计占 4 0 0 0万以上 .发展有效的丙型肝炎疫苗刻不容缓 .构建了 3段串联重复的丙型肝炎病毒多表位抗原基因 3pcx ,将其克隆到His融合表达载体 pET 2 8b(+)上后在大肠杆菌BL2 1(DE3)中得到了高效表达 .3PCX经Ni2 + NTA agarose柱纯化后免疫BALB/c小鼠 ,诱发了高水平的抗体免疫应答 . Hepatitis C virus(HCV) is the major causative agent of non-A, non-B hepatitis throughout the world. It is estimated that about 170 million people of the world have been infected with HCV and about 40 million of them are Chinese. There is a pressing need to develop an effective vaccine against HCV in order to combat this world wide public health threat. This work constructed a three tandem repeats of a mutlple-epitope antigen gene called 3 pcx and then cloned it into the expression vector pET-28b(+) to express it as hexahistine-tagged protein in Escherichia coli BL21(DE3). After purified by Ni^(2+)-NTA-agarose column, the protein was used to immunize BALB/c mice and it elicited strong antibodies responses.
