摘要
目的 :探讨外源基因在转基因动物中的遗传规律。方法 :将血管内皮细胞组织特异性表达人CD59基因转基因昆明种雄性小鼠 ,与普通昆明种雌性小鼠交配。Southern -blot杂交确定子一代外源基因整合阳性转基因小鼠。RT -PCR方法用于子一代转基因小鼠人CD59转录水平筛选。以流式细胞术检测人CD59基因在子一代转基因小鼠蛋白质水平表达。结果 :产子14只。7只外源基因整合阳性 ,其中雄性4只 ,雌性3只。转录水平检测发现3只出现所需的条带。继而对上述3只转录水平阳性的F1代小鼠白细胞行流式细胞术检测 ,白细胞膜表面人CD59表达均阳性。结论 :转基因动物的遗传规律复杂 。
Objective:To explore the rule of heredity in transgenic animals.Methods:Transˉgenic mouse tissue-specifically carrying human CD59on the vascular endothelium was mated with ordinary one.Southern blot analysis of genomic DNA was used to assess the presence of the transgene in the genome of the offspring,and expressions at mRNA and protein levesl were measured by RT-PCR and flow cytometry respectively.Results:Southern blot analysis showed that7of14offspring were transgenic.Human CD59gene was expressed on the surface of leuˉcocytes in3of the7hCD59-integrated offspring.Conclusion:The rule of heredity in transgenic animals is complicated.Gene Silencing may be a primary factor that some animals don't express the transgenes.
