硫喷妥钠抑制离体人单个核细胞分泌前炎症性细胞因子 被引量:6

摘要 目的 体外研究硫喷妥钠对离体人单个核细胞前炎症性细胞因子的影响。方法 抽取健康志愿者外周静脉血50ml,以密度梯度离心法提取其中单个核细胞,除阴性和阳性对照外,分别以100、400和1000μg/ml硫喷妥钠共同培养24h,收获前1h给予诱导物豆蔻酰佛波醇乙酯(PMA)或肿瘤坏死因子α(TNF-α)。通过酶联免疫吸附实验或化学荧光免疫吸附实验分析并比较细胞因子含量。结果 100、400和1000μg/ml硫喷妥钠使PMA诱导的IL-2分别从(511±320)pg/ml减少至(207±244)、(41±24)和(22±12)pg/ml,使IL-6从(270±293)pg/ml减少到(104±101)、(29±38)和(14±16)pg/ml,而TNF-α诱导的IL-8由(118±37)pg/ml减少为(40±14)、(24±11)和(12±19)pg/ml,以上改变均有统计学差异。结论 硫喷妥钠在体外呈剂量依赖性抑制人单个核细胞分泌前炎症性细胞因子IL-2、IL-6和IL-8。 Objective An in vitro study was designed to evaluate the influence of thiopental on preinflammtory eytokines, IL-2, IL-6, IL-8, secreted by human peripheral mononuclear cells. Methods 50 ml peripheral venous blood was drawn from each healthy donor. Mononuclear cells were isolated by density gradient centrifugation. In addition to negative and positive control, the cells were treated with 100,400 or 1000μg/ml thiopental for 24 hours respectively. PMA or TNF-α was added one hour before harvesting the cells. Both the cells and their supernatant were collected. IL-2 and IL-6 were measured by enzyme-linked immunosorbent assay (ELISA). IL-8 was analyzed by chemiluminescent-linked immunosorbent assay. Results After co-incubated with 100,400 or 1000μg/ml thiopental,IL-2 reduced from (511±320) pg/ml to (207±244),(41±24) and (22±12)pg/ml; IL-6 reduced from (270±293)pg/ml to (104±101),(29±38) and (14±16)pg/ml; and IL-8 decreased from (118±37) pg/ml to (40±14), (24±11) and (12±19)pg/ml respectively. All the changes above were statistically significant. Conclusion Within clinically-relevant concentrations, thiopental may suppress the secretion of preinflammtory cytokines in a dose-dependent manner.
