STZ糖尿病大鼠视网膜病变模型的评价 被引量:13

摘要 目的 评价链脲佐菌素 (STZ)糖尿病大鼠视网膜病变动物模型。方法 建立糖尿病大鼠模型 ,随机分为正常对照组 (M ) ,糖尿病 1个月组 (M1)、3个月组 (M3 )、5个月组 (M5)。分别进行视网膜铺片、石蜡切片及透射电镜超微观察。结果 普通石腊切片 :M、M1未见异常改变 ,从M3 开始出现异常改变 ,以M5最明显。表现为视网膜水肿 ,尤其内核层。细胞排列紊乱 ,内核层血管周围水肿更明显 ,节细胞数量减少 ,静脉扩张、渗出 ,并有出血。视网膜消化铺片 :( 1)周细胞数量 :M1与M相比无差别 (P >0 0 5 ) ,而M3 及M5与M相比明显减少 (P <0 0 1)。 ( 2 )毛细血管形态 :以M5时病变最重 ,可见毛细血管栓塞 ,无细胞毛细血管及毛细血管无灌注。透射电镜观察 :M未见异常改变 ,从M1开始出现异常改变 ,以M5最明显。表现为基底膜节段性增厚、断裂缺失 ,内皮细胞肿胀变形 ,向管腔内指状突起 ,异染色质浓集居边 ,周细胞核异染色质浓集居边 ,线粒体肿胀变性 ,甚至呈空泡状 ,视细胞外节盘膜结构紊乱 ,内节线粒体肿胀。结论 STZ糖尿病大鼠视网膜病变可以作为人类糖尿病视网膜病变的背景期病理改变的模型。 ObjectiveTo observe the animal model of diabetic retinopathy indcuced by STZ in rat based on histology analysis.MethodsDiabetes was induced in 35 rats with a single intraperitoneal injection of streptozotocin (STZ).Experimental rats were divided into M 1(diabetic for 1 month) ,M 3(diabetic for 3 months) and M 5(5 months) groups,and other 10 rats were assigned to normal control group.The morphology of retinal paraffin slide,digest preparation and ultrastrucure were observed by transmission electron- microscope.ResultsDifferent degree of lesion was found in diabetic rats of M 3 group and the severest lesion in M 5 group,including retinal edema(especially intern granulose layer),disorder of cell structure,decrease of ganglionices number,retinal venae expansion,exuding and hemorrhage.There was no difference of numbers of pericytes in retinal digest preparation between the M 1 group and the control (P>0.05),but obviously decreased in M 3 and M 5 groups(P<0.01-0.001).The severest morphologic change of capillary was seen in M 5 group,including thrombus and loss irrigation of capillary.Different degree of ultrastructure change was seen in M 1 group,showing thickness of fragmentation or split of basement membrane,edema of endothelium cell and finger-like process to the capillary cavity,marginal heterochromatin in endothelium and pericyte,edema and degeneration of mitochondrion,disorder of the ultrastructure of discus membrane of external fragment and edema of mitochondrion of internal fragment of optic cell.No ultrastructure and general histology changes were found in the control group.ConclusionDiabetic retinopathy model in rat may represent background stage of diabetic retinopathy in human.
