大鼠骨髓基质细胞向神经细胞分化的体外诱导动态观察 被引量:3

摘要 目的 动态观察体外定向诱导大鼠骨髓基质细胞 (BMSCs)分化为神经细胞的时程变化。方法 采用Percoll Paque液 (1 0 73g/ml)分离BMSCs体外扩增培养后 ,加入丁羟茴醚 +二甲基亚砜诱导其分化为神经细胞 ,分别于 1、2、3、4h后光镜下观察形态变化及通过免疫组化和免疫荧光染色测定特异性神经元烯醇化酶 (NSE)、神经丝蛋白 (NF)、胶质纤维酸性蛋白 (GFAP)阳性细胞数。结果 BMSCs在加入诱导剂后 1h出现胞体收缩 ,突起伸出 ,有不同程度NSE、NF、GFAP表达 ,3h后达高峰。结论 丁羟茴醚 +二甲基亚砜在一定时间内是一种较好的体外诱导剂。 Objective To observe the change of the induction of the differentiation of rat bone marrow stromal cells into neurons.Methods BMSCs were separated with Percoll-Paque reagent,cultured and expanded in vitro.Then BMSCs were induced with dimethyl-sulfoxide(DMSO)/butylated hydroxyanisole(BHA).We observed neuron morphology and detected the number of NSE+,NF+,GFAP+ cell with immunocytochemistry and immunocytofluorescence in 1,2,3h.Results DMSO/BHA can cause cells express neurons characteristic markers.After 3h the positive cells exhibited value peak.Conclusion DMSO/BHA is an effective induction material which can induce rBMSCs to differentiate into neurons.
