Effect of Al_2O_3 on Membrane Structure of Rat PAM

摘要 将大鼠肺泡巨噬细胞(PAM)与Al_2O_3共同体外培养。透射电镜下可见PAM 变形,伪足消失,溶酶体数量早期增条,后期减少,胞质内出现大空泡,核固缩,胞膜缺损,最后细胞崩解。培养基中酸性磷酸酶及乳酸脱氢酶显著增高。揭示Al_2O_3可造成PAM 膜性结构的明显损伤。 Pulmonary alveolar macrophage(PAM)of male Wistar rat were culturedwith Al_2O_3.Under transmission electron microcope,PAM was found to bechanged in shape with disappearance of pseudopodia.Lysosome increased atfirst,but decreased later and appeared as coarse particles.Large physalidesand pyknosis of cell nucleus can be seen.Ultimately,cell membrans were dam-aged with disintegration of PAM,and an increase of ACP and LDH activitiesin the culture medium.It suggested that Al_2O_3 might have a toxic effect onPAM,and be able to elicit apparent damage on its membrane structure.
