摘要 目的 研究血管内皮生长因子 (vesselendothelialgrowthfactor,VEGF)和色素上皮衍生因子 ( pigmentepitheliumderivedfactor,PEDF)在实验性小鼠脉络膜新生血管(choroidalneovascularization ,CNV)组织中的表达情况。探讨二者在CNV形成过程中所起的作用。方法 用半导体激光诱导小鼠CNV模型。分别于激光后 1、3、7d、2和 3周时取出眼球 ,采用原位杂交方法检测CNV组织中VEGF和PEDFmRNA的表达情况。结果 VEGF和PEDFmRNA在激光诱导的小鼠CNV组织形成过程中均有显著表达。激光光凝早期二者的表达均增高 ,但VEGFmRNA的表达升高更显著。激光照射后 3和 7d时 ,VEGFmRNA的表达即达到高峰 ,阳性率分别为 2 6 0 5 %和 2 7 92 % ,而PEDFmRNA的阳性表达率分别为 2 1 13%和 2 3 5 5 % 2周时 ,VEGFmRNA表达开始下降 ,约为2 3 95 % ,而PEDFmRNA的表达则达到高峰 ,为 2 9 19% 光凝后 3周时 ,二者的表达均下降 ,但PEDFmRNA的表达仍高于VEGFmRNA的表达 ,分别为 2 4 87%和 2 1 93% 结论 VEGF和PEDFmRNA明显表达于实验性小鼠CNV组织中。 2者表达失衡可能在CNV的形成过程中起到调控作用。 Objective To study the expressions of vessel endothelial growth factor(VEGF) and pigment epithelium derived factor(PEDF) in experimental choroidal neovascularization (CNV) tissues in mouse and their functions in the development of CNV.Methods CNV mouse models were induced by diode laser and the eyeballs were taken out at the 1st, 3rd, 7th day, 2nd and 3rd week.The expression of PEDF and VEGF mRNA in CNV tissues was detected by in situ hybridization.Results PEDF and VEGF mRNA were both significantly expressed in experimental mouse CNV tissues induced by diode laser.Their expression both increased during the earlier period after photocoagulation,but the expression of VEGF mRNA increased more significantly.Three and 7 days after laser treatment, the expression of VEGF mRNA got the highest,which positive expression ratio was 26.05% and 27.92% respectively,and the positive expression ratio of PEDF mRNA was 21.13% and 23 55%.Two weeks after photocoagulation, the expression of VEGF began to decrease,which was about 23 95%,while the expression of PEDF achieved the peak,which about 29 19%.At the 3rd week,they both decreased,but the expression of PEDF mRNA was still higher than VEGF,which was 24 87% and 21.93% respectively.Conclusion VEGF and PEDF mRNA are significantly expressed in experimental CNV tissues;The imbalance between their expressions may play an important role in the development of CNV.
