脊髓半切损伤后脊髓与大脑部分核团c-fos蛋白表达的实验研究 被引量:3

摘要 目的 :探索脊髓半切损伤后 ,脊髓、下丘脑室旁核 (PVN)和视上核 (SON)的c -fos蛋白表达的规律。方法 :将 5 6只大鼠随机分为脊髓损伤组 2 4只、假手术组 2 4只和正常组 6只 ,损伤组行左侧半脊髓横断术 ,损伤后 1,6,12 ,2 4h取脊髓、PVN和SON行冰冻切片 ,免疫组化法标记c -fos阳性细胞。结果 :脊髓半切损伤后 ,各时间点损伤侧脊髓灰质中有明显的c -fos阳性表达。PVN和SON在损伤后各时间点均有c -fos表达阳性细胞的增加 ,与假手术组、正常组比较 ,差异有显著性意义 (P <0 .0 5 )。结论 :脊髓损伤后脊髓、PVN和SON可产生明显反应 ,PVN和SON以不同的反应特点影响着神经递质的改变。 Objective:To explore the change of c-fos protein expression in spinal cord?hypothalamic paraventricular nucleus(PVN) and supraoptic nucleus(SON) after spinal cord hemisection.Methods:56 adult SD rats were randomly divided into injury group(n=24, Group A)?false operation group(n=24, Group B) and control group(n=6), spinal cord hemisection was performed in the left side. 1,6,12,24h after injury, spinal cord?hypothalamic PVN and SON were taken to crystal section for immunohistochemistry to detect c-fos positive cells.Results:c-fos was significantly expressed in gray matter of injuried side spinal cord?hypothalamic PVN and SON 1,6,12,24h after injury, compared with Group B and control group(P<0.05). Conclusions:The present findings suggest that spinal cord injury induces significant reaction in related nuclei in brain and hypothalamic PVN and SON affect the changes of neurotransmitter differently
